Objective To provide the evidence for the application of DYS413(YCAIII) in forensic science and population genetics by investigating its polymorphism in the Han population lived in Shanxi province and Mongolians lived in Inner Mongolia Autonomous Region, China. Method To collect respectively 180 unrelated males' venous blood 500ul, who lived in Shanxi province, 120 unrelated mongolians' venous blood 500ul, who lived in the Inner Mongolia Autonomous Region, and the blood is anticoagulant with EDTA, then to extract DNA by using the method of phenol-chloroform after ingested by proteinase K at 56 and amplify the DYS413 site by usingPCR. The products were separated with 6% non-denaturedpolyacrylamide/bisacrylamide gels(19:1) electrophore-sis and stained with l%oAgNO;).Result There are total 22 different haplotypes inShanxi Han population and 30 different haplotypes inMongolians, and the frequencies are respectively from0. 0056 to 0. 2056 and from 0. 0083 to 0. 1167, and the twoHD(haplotype diversity) are respectively 0.90248 and0. 94054.Conclusion It shows the DYS413 is high valuable in theapplication of forensic Medicine and populationgenetics.
