The purpose of this study was to assess the effects of exercise and oral glutamiiie supplementation on the concentration of the total protein and IGFs in muscles.32 male SD rats were randomly divided into three groups, i. e. control group, exercise combined with placebo supplementation group and exercise combined with glutamine supplementation group. JEhen exercise combined with placebo supplementation group and exercise combined with glutamine supplementation group were again divided into 2 day group and 5 day group. The muscle samples were taken 24 hours after the last exercise. Muscle protein concentration was determined from whole muscle homogenate by using the biuret method. The concentration of IGF- I and IGF- II were detected from whole muscle homogenate by RIA.The results showed that a significant decrease was found in the concentration of muscle protein at exercise. After 2 day and 5 day of exercise the concentration of the total muscle decreased by 19.36% (P<0.05) and 14.90% (P<0.05). An acute increase of the concentration of IGFs in the muscle. After 2 day and 5 day of exercise the concentration of IGF- I in the muscle increased by 36.65% (P<0.01) and 24.72% (P<0.05). After 2 day and 5 day of exercise the concentration of IGF-II in the muscle increased by 86.36% (P<0.01) and 63.64% (P<0.05). Oral glutamine supplementation have a great influence in the range of exercise-induced decline of the muscle total protein concentration and the exercise-induced rise of the muscle IGFs. Compared to exercise combined with placebo supplementation group. After 2 day and 5 day of exercise the concentration of the total muscle increased by 14.68% (P<0.05) and 24.43% (P<0.05). After 2 day and 5 day of exercise the concentration of IGF- I in the muscle decreased by 11.71% (P<0.01) and 19.93% (P<0.05). Alter 2 day and 5 day of exercise the concentration of IGF- II in the muscle decreased by 17.89% (P<0.01) and 28.33% (P<0.05)..It would appear that exercise could decrease the concentration of total muscle protein and result in muscle degradation, then stimulate the secretion of IGFs in skeletal musclewhich attend to the proliferation and differentiation of muscles. When the exercise went on, the skeletal muscle could adapt to the exercise, and the concentration of total muscle protein hasn't decreased, the secretion of IGFs in muscle went back to the level before the training. Additionally, it would suggested that supplying glutamine play an important role in protecting the muscle issues, improving the muscle function, increasing the speed of recovery. And enhancing muscle synthesis, decreasing muscle break down. In the same conditions, the muscle can endure the more intensity. Shorten the process of the muscle adaptation to exercise.
