Obejective: To establish the SD rats model that conforms to II diabetes characteristics, and investigate the effect of exercise and puererin on antioxidant and insulin resistance in diabetic rats,and discuss the scientific of II diabetic theories by aerobic exercise and medicine supplemented.Methods:①70 healthy male SD rats aged 8 weeks were selected and randomly divided into 2 groups: normal control group with 16 rats, experimental group with 54 rats.②Experimental groups were fed for 4 weeks by the hyper-lipid feedstuff and injected STZ (45mg/kg). Only rats with blood glucose levels > 14.7mmol/L and urine suger ++ ~ ++++ were selected as the experimental protocols. The normal control group was fed ordinarily, and injected 0.1 mmol/l citrate buffer at the same volume. After modeling, the DM rats were randomly divided into 4 groups: DM control group, DM exercise group, DM puerarin group, and DM exercise plus puerarin group.③The rats in the DM exercise group ran at the speed of 15 m per second for 60 minutes on the treadmill with 5°slope for 6 weeks. Puerarin group was intragastrically infused with puerarin by 500 mg/ kg body mass for 6 weeks; the DM exercise plus puerarin group was given puerarin infusion and treadmill running training. Each group was fed by ordinary feedstuff.④before and after intervene, the changes in T-AOC, CAT, GSH-PX, CAT activity, GSH, MDA contents, body mass, blood glucose, insulin and Insulin sensitivity index were measured.Results: The method was proved practical to establish the SD rats model into type II diabetic characteristics by fed hyper-lipid feedstuff and injected with STZ. After experiment, The level of T-AOC, CAT, GSH-PX, CAT activity and insulin of intervene diabetic groups were significantly higher (P<0.05),while MDA contents and blood glucose were very lower (P<0.01).Compared with the DM control group,The level of T-AOC,SOD,GSH,GSH-Px of the DM exercise plus puerarin group was significantly higher(P<0.01),the MDA contents and blood glucose declined significantly(P<0.01).Compared with the normal contrl group,the level of ISN of the DM exercise plus puerarin group had remarkable difference (P<0.05), while the level of FBG,T-AOC,SOD,CAT,GSH,GSH-Px and MDA had no obvious changes(P>0.05) . Therefore, we can make a conclusion that aerobic exercise plus puerarin could enhance serum antioxidant of diabetic rats, reduce lipid perOxidative damage and improve the level of blood glucose and eliminate the insulin resistance. Conclusion:(1) the SD rats were modeled into type II diabetes mellitus by fed hyper-lipid feedstuff for 4 weeks and injected with streptozotocin, It is similar to human being. And the DM model can offer the condition to develop the extensively research in laboratory.(2) Aerobic exercise plus puerarin intervention can improve the insulin resistance phenomenon on the type II diabetic mouse.(3) Exercise and puerarin could enhance serum antioxidant of diabetic rats, reduce lipid perOxidative damage and improve the level of blood glucose and insulin. Exercise combined puerarin showed best outcome among all experimental groups. The combination of multifarious efficient treatments would obtain a satisfactory healing for diabetes.
