| The concept of pre-hypertension involving blood pressure at 120/139~80/89 mmHg, was put forward by the JNC-7 in 2003, which is consistent with the concept of "normal high blood pressure" in our country’s latest definition of hypertension in 2010. Due to the tight, fast pace of work, high blood pressure in the early stage caused by the chronic psychological stress is increased year by year especially among adolescents. Electro-acupuncture therapy has been widely used in many hypertension-related cardiovascular diseases. Researchs showen that the acupuncture rugulates blood pressure and cardiovascular function by improveing NO/NOS system, sympathetic nervous system and vascular endothelial function.PCR technology has the characteristics of high sensitivity, small sample size, high specificity and precision, which is suitable for the study of micro biological changes in the early stage of hypertension.ObjectiveThis study focused on the subject of electro-acupuncture to regulate blood pressure and protect vascular endothelial function in rats with stress induced hypertension and under the guidance of "prevention before disease"in traditional Chinese thought to observe the effect of electro-acupuncture on serum Ach, NPY content and the CXCR-2, MMP-9 and MCP-1 gene expression. We will reveal electro-acupuncture’s effect on blood pressure lowing and vascular endothelium protecting mechanism.MethodsA total of 27 male rats were randomly divided into blank control, model, electro-acupuncture groups (n=9 in each group).The stress pre-hypertension model was established by giving the rats with noise and electric shock stimulation (paw),twice a day for 14days.Electro-acupunc-ture stimulation was applied to bilateral "Taichong, Quchi" (LR3, LI11) for 20 min.The treatment started as soon as the stress stimulation set out. Systolic blood pressure of the rat’tail was detected with non-invasive method and Serum contents of NPY and Ach were measured by using ELISA method and CXCR-2ã€MMP-9 and MCP-1 gene expression with PCR.ResultsCompared with the control group, the systolic blood pressure was significantly higher in the model group after 11 day’s stress stimulation(P<0.01),while the contents of serum NPY were markedly up-regulated(P<0.01) and the contents of serum Ach were markedly down-regulated(P< 0.01). With the elctro-acupuncture interference,the blood pressure was lower((P<0.01, P<0.05), the contents of serum NPY were markedly dow-regulated(P<0.01) while the contents of serum Ach were markedly up-regulated(P<0.01)in contrast to the model group.Compared with the control group,the expression of CXCR-2(P<0.01)ã€MMP-9(P<0.01) and MCP-1 (P<0.01) was remarkably higher in model group.While CXCR-2(P<0.01)〠MMP-9(P< 0.01) and MCP-1 (P<0.01) expression was significantly lower in the elctro-acupuncture control group compared with the model group.Conclusion1. EA at "Taichong and Quchi" has a significant effect to suppress the elevated blood pressure in stress induced PHT rats.2. EA at "Taichong and Quchi"can affect the contents of serum NPY and Ach.3. CXCR-2ã€MMP-9 and MCP-1 gene expression may be involved in EA’s anti-hypertension mechanism. |
