Aims:This study choose hypertension related vascular dementia (vascular endothelial growth factor,VD) model rats as research objects, to discuss changes between EA and acupoint application on blood pressure, cognitive function and vascular endothelial growth factor (VEGF) of model rats before and after treatment and provide more adequate scientific basis for EA and acupoint application in treating for vascular dementia.Methods:To choose Wistar rats as research objects. To set up hypertension related vascular dementia models by the high salt diet and 2-VO method. The rat tail artery blood pressure measuring instrument and Morris water maze were used to screen the model. Model rats were randomly divided into model group, EA group and acupoint application group, giving EA therapy and acupoint application therapy. To monitor rat tail artery blood pressure measurements in early, middle and late stages of the treatment on three groups, and to use the Morris water maze test the cognitive and learning ability, and analysis the difference of rat vascular endothelial growth factor (VEGF) by using enzyme linked immunosorbent assay (enzyme-linked immunosorbent assay,ELISA) technique after the treatment.Results:1.The blank group compared with the other three groups.after the model maked,the other groups of blood pressure.the abilty of learning and memory and VEGF are statistically significant(P< 0.05).2. Blood PressureComparing the EA group and the model group, blood pressure of EA group decreased significantly after treatment(P< 0.05), The longer treatment time is, the effect of rats'blood pressure is more significant; Comparing the acupoint application group and the model group, blood pressure of acupoint application group decreased significantly (P< 0.05), and the longer treatment time is, the effect of rats'blood pressure is more significant. The time factor affecting blood pressure value of the change in the point of the difference was statistically significant (P< 0.05); in different time and groups, the blood pressure value all had interactive effect (P< 0.05), the effect of time factor varies with the grouping.3. Learning and memoryPositioning navigation test:comparing EA group and the model group, escape latency of rats after EA treatment was significantly shortened, the differences have statistical significance (P< 0.05); comparing acupoint application group and model group, escape latency of rats after acupoint application changed little, no statistically significant differences (P> 0.05). The time factor affecting escape latency of the change in the point of the difference was statistically significant (P< 0.05); in different time and groups, the escape latency all had interactive effect (P< 0.05), the effect of time factor varies with the grouping.Spatial probe test:comparing EA group and the model group, frequency rats after EA treatment going across the primary platform increased significantly, the differences have statistical significance (P< 0.01); comparing acupoint application group and model group, frequency rats after EA treatment going across the primary platform changed little, showing no significant statistical significance (P> 0.05). The time factor affecting frequency rats of the change in the point of the difference was statistically significant (P< 0.05); in different time and groups, the frequency rats all had interactive effect (P< 0.05), the effect of time factor varies with the grouping.4. Enzyme linked immunosorbent assay analysis testComparing EA group and the model group, the VEGF concentration after EA treatment was significantly increased, the effect of differences between the two groups have statistical significance (P< 0.01); comparing acupoint application group and model group, the VEGF concentration after acupoint application increased, but differences in the effect of no statistical significance (P> 0.05); comparing EA group and acupoint application group, the VEGF concentration after EA treatment was significantly higher, the effect of differences between the two groups has statistical significance (P< 0.01).Conclusion:1.EA and acupoint application can both lower blood pressure and the effect is more obvious treatment when the treatment time is longer;2.In behavioral tests, the ability of learning and memory in the EA group compared with the model group had greatly improved after a certain time of treatment, although acupoint application group improved slightly, but it did not have a big impact;3.In the enzyme linked immunosorbent analysis test, the VEGF concentration of EA group improved evident and acupoint application group have certain ascend, that two kinds of therapies have certain effect to rats on angiogenesis and nerve protection.
