Objective:To study the variation of the wound healing rate and relevant factors in the process of wound healing when external apply traditional Chinese medicine Yinhuang San in diabetic ulcers of rats,and then explore the mechanisms of external application of traditional Chinese medicine Yinhuang San in diabetes ulcer healing of rats.Methods:Raise 141 male SD rats of 80-100g with high fat and sugar a month then intraperitoneal injection STZ 35mg/kg to prepare the model of diabetic rats, and observe FBG a week. The 141 diabetic rats cut modeling area of skin deep to fascia under sterile conditions. The establishment of diabetic ulcers model rats was randomly divided into model group, western medicine group and TCM group, each 47, respectively, corresponding to the drug.We drawn materials when the 5th day,the 10th day,the 15th day, using immunohistochemistry to detect bFGF、PDGF、TGF-β、AGEs、 AGER、TNF-α、IL-1α levels in the wound granulation tissue;we took photos everyday when the wound were dressed,using Image 1.46r software to measure the wound area of the 5th day,the 10th day,the 15th day,and then calculated the wound healing rate. Use SPSS 20.0 software to statistically analyze immunohistochemical results of wound granulation tissue and wound healing rate.Results:Compared with the model group,TCM group and the western medicine group can increase the rate of wound healing significantly(P<0.01), but TCM group is superior to western medicine group(P<0.05).Compared with the model group, TCM group and the western medicine group can improve bFGF levels of granulation tissue in the whole process of wound repair(P<0.01, P<0.05);in the early and middle of wound repair, bFGF levels of granulation tissue are the same in TCM group and the western medicine group(P>0.05), in the late of wound repair,TCM group below the western medicine group(P<0.05).Compared with the model group,PDGF levels of TCM group’s granulation tissue are improved significantly in the late of wound repair(P<0.01); Compared with the model group,PDGF levels of the western medicine group’s granulation tissue are improved significantly in the early of wound repair (P<0.01); Compared with the western medicine group, PDGF levels of TCM group’s granulation tissue are below significantly in the early of wound repair (P< 0.01),same in the middle of wound repair (P>0.05), improved in the late of wound repair (P <0.05).In the whole process of wound repair,TGF-β levels of three group’s granulation tissue are the same(P> 0.05).Compared with the model group,AGEs levels of TCM group’s granulation tissue are below significantly in the whole process of wound repair (P<0.01); compared with the model group,AGEs levels of the western medicine group’s granulation tissue are below significantly in the early and middle of wound repair (P<0.01); compared with the western medicine group, AGEs levels of TCM group’s granulation tissue are below significantly in the early and late of wound repair (P<0.01).Compared with the model group and the western medicine group,AGER levels of TCM group’s granulation tissue are below significantly in the whole process of wound repair (P<0.01);compared with the model group, AGER levels of the western medicine group’s granulation tissue are below significantly in the early of wound repair (P<0.01).Compared with western medicine group,TNF-α levels of TCM group’s granulation tissue are below significantly in the early and middle process of wound repair (P<0.01); compared with the model group, TNF-a levels of the western medicine group’s granulation tissue are below significantly in the whole process of wound repair (P<0.01); TNF-a levels of TCM group’s and the western medicine group’s granulation tissue are the same(P>0.05).Compared with the model group,IL-1α levels of TCM group’s granulation tissue are below in the early and late of wound repair (P<0.05); compared with the model group,IL-la levels of the western medicine group’s granulation tissue are below significantly in the late of wound repair (P<0.05);compared with the western medicine group, IL-la levels of TCM group are below in the whole process of wound repair,but are below significantly in the early and middle of wound repair (P<0.05).Conclusions:TCM and western medicine can increase the rate of wound healing significantly, speed up the wound healing and shorten the wound healing time. But TCM Yinhuang San’s clinical effect is better. The mechanisms may be:stimulus wound repair cells’s proliferation and differentiation, promote angiogenesis and collagen synthesis,improve the inflammatory condition of the wound,and reduce AGEs and AGER levels of the wound.
