Research Targets:Dioscoreae Althaeoides is one of the most commonly used traditional Chinese medicines, and its quality directly affect clinical efficacy. Although the specification and grade have been classified by "76 kinds of medicinal products specifications", with the change of ecological environment and modern growing conditions, the existing standard specification level has clearly no longer applicable to the market demand. It is necessary to establish a new classification standard of Dioscoreae Althaeoides.Research Methods:The Delphi method, which can unify different experts’sensory identification experience of Dioscoreae Althaeoides from different areas and different seasons throughing the questionnaire survey, was used in the study to determine the main indicators of the evaluation of medicinal products specifications, and analyze the accuracy and the scientific nature of the sensory evaluation.Observing appearance, polarizing light microscopy and infrared spectroscopy identification were used to evaluate different levels of quantization of Dioscoreae Althaeoides and achieve the further identification. Finally, the main indexes of different levels of Dioscoreae Althaeoides were determined by chemical method, and the differences between different levels were distinguished.The study was divided into four parts:The first part, the main evaluation indexes were determined and the specification and grade were divided by Delphi method.The second part, the important evaluation indexes were clared and quantified by observing appearance, polarized light microscopy. The third part, the further identification of different Dioscoreae Althaeoides was achieved by infrared spectroscopy identification. The forth part, the content of the composition of the main indicators were mensurated by chemical determination.Research Results:The First Part:The specification and grade of Dioscoreae Althaeoides were evaluated by Delphi method.The results show:The method of sensory identification was objective and reproducible, and to a certain extent, the evaluation results ware scientific and accurate. The main evaluation indexes included:appearance and profile features, combined with sheath diameter and fibrous roots, and the origin and variety were the most important factors that determine the size of Dioscoreae Althaeoides medicinal products. The Dioscoreae Althaeoides were dieved into two specifications and there levels.The iron Dioscoreae Althaeoides were Henan shatu tiegun Dioscoreae Althaeoides, Shandong tiegun Dioscoreae Althaeoides and Henan lutu tiegun Dioscoreae Althaeoides. The non-iron Dioscoreae Althaeoides included Dioscoreae Althaeoides from Henan, Hebei and Shanxi.The Second Part:The appearance characters and polarizing microscopic identificationThe results show:The main difference between different herbs in appearance shape were skin color, diameter, and section characteristics of roots; in the microscopic characteristics mainly were the distribution density and the size of calcium oxalate crystal bundle, and the distribution density of the vascular bundle.The Third Part:Infrared spectrum identificationThe results show:The infrared spectrum of yam was similar to that of starch, which showed that the yam contains a lot of starch. The yams which one dimensional map were similar can be identified by the three levels of identification. The main difference between different herbs was the different peak intensity in the region of 1250 cm-1 to 900 cm-1, and the different peak shape 1800 cm-1 to 1300 cm-1. At the same time,0.9900 as threshold, can be distinguished the yam from other areas and Henan in the similarity comparison of infrared spectra.The Fourth Part:Determination of main index componentsThe results show:The polysaccharide content was quite different between different herbs, but the allantoin content had no significant difference. Shanxi Chang yam content the highest polysaccharide, was 18.28%+ 0.48. Followed by Hebei origin of yam and Henan’s was lower, which was 10.64+ 1.07. Shandong Iron yam polysaccharide and allantoin content were the lowest,8.42% and 0.77%.Research Conclusion:In summary, the different Dioscoreae Althaeoides were dieved into two specifications and there levels preliminarily.The iron Dioscoreae Althaeoides were Henan shatu tiegun Dioscoreae Althaeoides, Shandong tiegun Dioscoreae Althaeoides and Henan lutu tiegun Dioscoreae Althaeoides. The non-iron Dioscoreae Althaeoides included Dioscoreae Althaeoides from Henan, Hebei and Shanxi. At the same time, there were significant differences in different specifications and levels Dioscoreae Althaeoides in appearance, polarized light microscopy characteristics, infrared identification. But there was some differences betwent the main indicators ingredient content and assessment results,which need certain further experimental study.
