Purpose:The purpose of this experiment,based on sciatic nerve crush injury model of rat,is to analysis and compare the effects of different frequencies(2Hz,100Hz) of electro-acupuncture needling Huantiao point,which is highly related to sciatic nerve,in the treatment.From the behavior of the sciatic nerve function index, nerve HE staining proved that acupuncture treatment of sciatic nerve injury is effective in inhibition of nuclear factor kappa B signal transduction pathway in perspective to inflammatory factor IL-1β,IL-6,TNF-α index and evaluate the inhibitory effect of acupuncture on inflammatory reaction and nerve injury.Based on the index to analysis the different frequencies of electro-acupuncture on sciatic nerve injury of inflammation inhibition and nerve repair mechanism.Material and method:We choose 48 Specific Pathogen Free(SPF) Sprague Dawley(SD) rats, male and female,weight 300 ± 50 g,7 to 9 weeks of age,half male and half female rats.After buying,fed in the animal experiment center of Liaoning University of Traditional Chinese Medicine.After feeding for 1 weeks,48 Sprague Dawley rats were randomly divided into control group,model group,2Hz group.100 Hz group,12 rats in each group.Control group:no treatment, normal feeding, keeping other conditions unchanged; animal model group:after modeling success,keeping other conditions unchanged,the normal diet;2Hz group:rats were at the modeling 8th day acupuncture intervention,acupuncturing Huantiao point to neural stem,rats muscle tremor,toe jitter,connection acupuncture device,frequency selection2 Hz,wave type continuous wave,current of 1m A,intensity of acupuncture site mild local tic as the standard,treatment time 15 min,1 /d,continuous treatment of 14d;in 100 Hz group,electro-acupuncture frequency is 100 Hz,everything is made with the 2Hz group.At the modeling 8th day(before the acupuncture treatment)and at the end of the treatment(modeling22th day),to take respectively in the model group,2Hz group and 100 Hz group rat sciatic nerve function index detection,evaluation of the rats in the recovery of neuromuscular and motor function before and after treatment.after the end of treatment(modeling twenty-second days), the materials were drawn,the rats were killed by anesthetic overdose,including injury,on the sciatic nerve tissue of each 2mm and L4-5 segment of the spinal cord,HE staining,immunohistochemistry staining was used to detect the damaged nerve inflammatory factor TNF-α,IL-1β,IL-6 content;by Western Blot expression level was detected in the spinal cord of Nuclear Factor kappa B protein.Finally,the results were statistically analyzed.Results:1. The SFI index of rats in each group show:At the modeling 8th day(before the acupuncture treatment),the model group,2Hz group and 100 Hz group SFI index were significantly lower,and the difference was not statistically(p>0.05);at the modeling 22 thday,after 14d’s treatment,2 treatment group rats SFI index was significantly higher than that in the model group,the difference was statistically significant(p<0.01),and the difference between the two was statistically significant(P<0.01),it can be seen that electro-acupuncture treatment can promote the recovery of motor function in rats,the 2Hz group is better than the 100 Hz group.2. The sciatic nerve of rats Hematoxylin-eosin staining show:Control group axons,Schwann cells and nerve fibers arranged orderly,continuous;model group’s nerve fibers were disordered,Schwann cells increased,swollen axons,myelin disintegration;electro-acupuncture 2Hz group is better than the model group,Schwann cells decreased,nerve fibers gradually and continuously,axon growth is good;electro-acupuncture of 100 Hz group’s nerve fiber disorder degree alleviated as compared with model group,Schwann cells decreased,the number of nerve fibers increased.From the observation of pathological images,electro- acupuncture group 2Hz sciatic nerve recovery degree is better than 100 Hz group.3. TNF-α, IL-1β, IL-6 content in the injured nerve of rats in each group immunohistochemical method results show:Immunohistochemical method was used to detect the damaged nerves of TNF- α,IL-1β,IL-6 were expression brown, mainly in the cytoplasm, no staining was negative expression.Compared with the control group,model group,TNF-α,IL-1β,IL-6 expression was significantly increased,the difference is statistically significant(P<0.05);compared with the model group,2 EA group TNF-α,IL-1β,IL-6 expression was significantly reduced,the difference is statistically significant(P<0.05);compared with the 2Hz group,high positive expression of 100 Hz group TNF-α,IL-1β,IL-6,the difference is statistically significant(P<0.05).4. In spinal cord of rats in each group of NF-κB protein expression results show:To compare with the control group,the other three groups in the rat spinal cord Nuclear Factor kappa B protein expression was significantly higher, the difference is statistically significance(P<0.05);compared with the model group,2Hz group,100 Hz group rat spinal cord Nuclear Factor kappa B protein expression is significantly lower, the difference is statistically significance(P<0.05);compared with the 2Hz group,100 Hz group rat spinal cord Nuclear Factor kappa B protein expression is significantly higher, the difference is statistically significance(P<0.05).Conclusion:1. Electro-acupuncture treatment can promote the repair of injured sciatic nerve,promote the recovery of motor function in rats with sciatic nerve injury,and 2Hz is better than100Hz;2. Electro-acupuncture treatment can effectively inhibit the inflammatory response and promote the repair of damaged nerves,and 2Hz is better than 100Hz;3. Electro-acupuncture treatment can effectively inhibit the expression of Nuclear Factor kappa B signaling pathway, reduce the inflammatory factor TNF-α, IL-1β, IL-6secretion,in favor of nerve regeneration and repair,and 2Hz is better than 100Hz;4. The effective inhibition of the expression of Nuclear Factor kappa B signaling pathway can effectively inhibit the expression of inflammatory factors,like TNF-α,IL-1β,IL-6 secretion,effectively inhibit the inflammatory response,it may be one of the mechanisms of acupuncture to promote the recovery of peripheral nerve injury.
