| Purpose:In this study, SPF Wistar male healthy rats were used as the research object, Acute Pancreatitis(AP) model was established by intraperitoneal injection of L- arginine in rats.Salvia miltiorrhiza group was injected with Danshen injection at different time before and after the model.To observe the effect of Salvia miltiorrhiza injection on serum tumor necrosis factor alpha and serum amylase activity in rats with acute pancreatitis.so as to evaluate the therapeutic effect of Salvia miltiorrhiza injection on acute pancreatitis.To further explore the mechanism of Danshen injection in prevention of acute pancreatitis.Material and method:Will rats aged 4-5 weeks sixty clean level Wistar healthy male rats,weighing 280± 20 g, number according to the random number table method,divided into 6 groups,respectively as the blank control group, model group and Danshen group(1, 2, 3, 4 groups),10 rats / group.According to Mizunuma T[1]ã€Yang Ping[2] literrature describes methods.The acute pancreatitis model was induced by intraperitoneal injection of 0.574mmol/L(10%) L- arginine solution in rats.The blank control group was given the same amount of normal saline.Salvia miltiorrhiza group at different time(before modeling, 30 min modeling, after modeling, 30 60min) intravenous injection of Salvia miltiorrhiza.2h after modeling, the rats received intraperitoneal injection of 10% chloral hydrate anesthesia,abdominal aortic blood vessel 3ml/per,ELISA method was used to measure the serum amylase and TNF- alpha in rats,and to evaluate the therapeutic effect of Salvia miltiorrhiza injection on acute pancreatitis.Results:1.The serum amylase levelAs for comparison with the control group, serum amylase level of rats in the model group and salvia groups is obviously higher than the control group(P<0.01). Compared with the model group, serum amylase level of rats in the salvia groups is obviously lower than the model group(P<0.01). Comparison between salvia groups: as for comparison between salvia group 1 and salvia group 2, serum amylase level of salvia group 1 is obviously lower than salvia group 2(P<0.05); as for comparison between salvia group 1 and salvia group 3, serum amylase level of salvia group 1 is obviously lower than salvia group 3(P<0.05); as for comparison between salvia group 1 and salvia group 4, serum amylase level of salvia group 1 is obviously lower than salvia group 4(P<0.05); as for comparison between salvia group 2 and salvia group 3, serum amylase level of salvia group 2 is obviously lower than salvia group 3(P<0.05); as for comparison between salvia group 2 and salvia group 4, serum amylase level of salvia group 2 is obviously lower than salvia group 4(P<0.05); as for comparison between salvia group 3 and salvia group 4, serum amylase level of salvia group 3 is obviously lower than salvia group 4(P<0.05)2.The TNF alpha levelAs for comparison with the control group, TNF-αlevel of rats in the model group and salvia groups is obviously higher than the control group(p<0.01). Compared with the model group, TNF-α level of rats in salvia groups is obviously higher the model group(P<0.01). As for comparison between salvia group 1 and salvia group 2, TNF-αlevel of salvia group 1 is obviously lower than salvia group 2(P<0.05). As for comparison between salvia group 1 and salvia group 3, TNF-αlevel of salvia group 1 is obviously lower than salvia group 3(P<0.05). As for comparison between salvia group 1 and salvia group 4, TNF-αlevel of salvia group 1 is obviously lower than salvia group 4(P<0.05). As for comparison between salvia group 2 and salvia group 3, TNF-αlevel of salvia group 2 is obviously lower than salvia group 3(P<0.05). As for comparison between salvia group 2 and salvia group 4, TNF-αlevel of salvia group 2 is obviously lower than salvia group 4(P<0.05). As for comparison between salvia group 3 and salvia group 4, TNF-αlevel of salvia group 3 is obviously lower than salvia group 4(P<0.05).Conclusion:1.Salvia miltiorrhiza injection can reduce the level of serum amylase in rats with acute pancreatitis.2.Salvia miltiorrhiza injection can reduce the content of serum tumor necrosis factor alpha in acute pancreatitis rats.3.The effect of Salvia miltiorrhiza injection was significantly different in different stages of acute pancreatitis4.the early application of Salvia miltiorrhiza injection on the treatment of acute pancreatitis is the best. |
