Purpose:From the view of epidermal growth factor and gastrin to discuss the mechanism of Xiaoyongkuidekang effect on the rats with gastric ulcer in type-acetic acid, which by utilizing molecular biology technique.Materials and methods:Male and female 80 Wistar rats(180±10)g were divided into 7 groups by random digit grouping method: normal group of 10(A), model group of 12(B), omeprazole in the treatment group of 12(C),Xiaoyongkuidekang treatment group of 12(D), heat-clearing and detoxifying treatment group of 12(E), strengthening the spleen and stomach treatment group of 10(F), 10(G) in the treatment group were treated with promoting pus discharge and tissue regeneration. Rats in all treatment groups divided into cages for 2 days, after the acetic acid preparation method of modeling, in the junction of sinus ventriculi and corpus ventriculi were injected by 100%glacial acetic acid 0.02 ml to build models of acute gastric ulcer, until the tunica serosa gastria appeared about 3mm white ulcerous ring and the stomach adapted back to abdominal cavity and sutured muscle and skin, then disinfected by iodophor. After successful modeling, the rats in each treatment group were intragastric administered for a total of 14 days. Each of A, B treatment group was given normal saline by daily according to the weight of 1.25ml/100 g of the rats, and the C treatment group was given omeprazole aqueous solution by the standard of1.04ml/100 g weight day by day; the rats in D treatment group according to the weight standard with 1.25ml/100 g were given intragastric administration by “Xiaoyongkuidekang”apozem. The rats in E, F and G treatment group according to the standard of the weight of1.25ml/100 g were given intragastric administration by apozem of Chinese Medicine, which for 14 days in totals. Two hours after the last administration, they were injected with anesthesia of 10% chloral hydrate in the abdomen, then treated with laparotomy, the ligation of pylorus and preventriculus, then picked out the whole stomach and placed in 10%formaldehyde solution fixed for 20 minutes. Then should be cut along the greater curvature of the stomach and paved after flushing with sterile saline. Cut the stomach parietal wall along the 3mm of edge of ulcer for inspect to be put to death. The quantitative polymerase chain reaction(Q-PCR) were used to detect the expression of epidermal growth factor m RNA in gastric tissue; ELISA method was used to detect the expression of gastrin in tissue. The results were statistically analyzed by the analysis of variance in SPSS19.0 software.Results:The expression of epidermal growth factor m RNA in B-G group decreased and there was a significant difference compared with group A(P < 0.05). After treatment, the expression of epidermal growth factor m RNA in C-G group was better than that of group B(P < 0.05);Compared with group C, the expression of epidermal growth factor m RNA in E-G group reduced(P < 0.05) and group D without no significant difference(P > 0.05). Compared with group D, the expression of epidermal growth factor m RNA in E-G group decreased(P < 0.05).Experimental results showed that the therapeutic outcome of Xiaoyongkuidekang decoction can improve the expression of egfmrna in acetic acid-induced gastric ulcer in rat gastric ulcer mucosa tissue, treatment effect is better than single treatment of a kind of treatment method.The expression of gastrin in B-G group decreased and there was a significant difference compared with group A(P < 0.05);After treatment, the expression of each treatment group was better than that of group B(P < 0.05);Compared with group C, the expression of gastrin in E-G group decreased(P < 0.05), and there was no significant difference compared with group D(P > 0.05);Compared with the D group, the expression of E-G group decreased( P <0.05). Experimental results show that the therapeutic outcome of Xiaoyongkuidekang can improve the content of acetic acid gastric ulcer in rat gastric ulcer mucosa of gas, treatment effect is superior to the use of a therapeutic approach in the treatment group.Conclusion:1. It confirmed the effectiveness of xiaoyongkuidekang for the healing of gastric ulcer in rats.2. "Xiaoyongkuidekang" was able to improve the relative expression of EGF m RNA and GAS content of gastric ulcer tissue in rats, shortened the healing period in order to improving the repair ability for impaired mucosal.
