Objectives: The gastric ulcer is a global common disease of the digestivesystem, which is formed because of helicobacter pylori, drugs and dietaryfactors, mental factors such as stress leading to excessive gastric acid outputand pepsin digesting their own stomach, causing gastric mucosa tissue damage.According to statistics, about10%of people in his life had the peptic ulceronce. Modern medical researches think that gastric ulcer is a typicalpsychosomatic disease. Bad emotional stress can affect gastrointestinalfunction which is the important factor causing the ulcer recurrence ofsymptoms or aggravated. As the experience of Professor Yang Muxiang who isthe famous old Chinese medicine expert, Weiyanyin has the effect of soothingliver and invigorating spleen, and harmonizing stomach and lowering adverseflow. This study through the observation of the effect of Weiyanyin on gastriculcer rats with the pathological morphology of gastric mucosa, epidermalgrowth factor (EGF), transforming growth factor (TGF), vascular endothelialgrowth factor (VEGF) effect and their mRNA expression, in order toinvestigate the soothing liver and invigorating spleen prescription promoteulcer healing and prevent the recurrence of the mechanism.Methods: With reference to the literature, copying gastric ulcer rats’model used the method of glacial acetic acid corrosion. The50depurator malewistar rats with the weights of200-220g were offered by the experimentalanimal center of Hebei Medical University. Adaptive feeding after one week,8rats were randomly selected as sham operation group, and the remaining42rats participated making the model.The third day after making the model, removing the dead rats, it neededto randomly select32rats from the survival rats,which were identified assuccessful models, then divid into model control group (called a model group for short), omeprazole positive control group (called a control group for short),WeiYanYin high-dose group (called a high-dose group for short) andWeiYanYin low-dose group (called a low-dose group for short), and each onetakes8rats. Each treatment group rats were to be filled the stomach with perkilogram of body weight to10ml of liquid, the high-dose group usedWeiYanYin decoction whose concentration was2.14g gruffs/ml, the low-dosegroup was WeiYanYin decoction whose concentration was1.07g gruffs/ml,and the control group used Omeprazole fluid whose concentration was0.368mg/ml.The sham operation group and the model group used thephysiological saline with per kilogram of body weight to10ml of liquid.Allabove groups were treated with drug or physiological saline once a day, andthe treatment lasted for14days.In the process of the drug treatment, it needed to observe each rat’s state ofmind, hair luster, eating and activity. After the last intragastric administration,all rats were stopped feeding for24h and decapitated, then were removed thestomaches by cutting the belly open with the aseptic technique. Theirstomaches were cut open along the greater curvature, washed with ice normalsaline, and firstly observed the general form of the gastric mucosa from themacro.Then measured the maximum length diameter and the verticalmaximum width diameter of the ulcer by sliding caliper, and calculate theulcer area and the inhibition ratio of ucler. Again from near the ulcer focal orthe healing place to cut gastric tissue, with4%paraformaldehyde fixed,conventional paraffin embedding, sectioning, HE staining, to observe the ulcerhealing under the light microscopy ulcer healing. Taking above-mentionedparaffin section, detected the expression of EGF、TGF-β1and VEGF by theImmunohistochemical method. Then removed the gastric tissue into glasshomogenate tube and added to prechill isotonic Na chloride which wereisovolumic, ice tray homogenate, detected the expression of EGFmRNA、TGFmRNA and VEGFmRNA by RT-PCR.The measurement data were expressed by modality of mean±standarddeviation(士S), which were statisticed by using the SPSS for Windows 16.0software processing. The single factor analysis of variance was used instatistical method, then “LSD” examination was used when comparingbetween two groups. The lever of significant difference was with P<0.05andP<0.01for standard.Results:1The general condition of every groupAfter the model making, the rats of the sham operation group were noobvious change, with dense and glossy hair, bright eyes, sensitive reaction,more action, normal food intake and granular manure. Other groups of ratswere in different degrees of hair disheveled, dim and yellow, and were lazy,unresponsive, eat less, cowered, defecate less and sticky. As the passage of thetime and the treatment of drug after the end of making model, theabove-mentioned symptoms of rats were gradually improving, among whichthe model group improved slower and each treatment groups improved morerapidly, especially the high group recovered fastest.2The pathomorphology changes of gastric mucosa among groups of rats2.1The observation on the gross morphologyThe gastric mucosait of the sham operation rats was reddish, had obviousfold, and had no ulcer, edema and hyperemiat on the surface. The gastric wallof the model group was concave ulcer with straight edge, round or oval.Thesurrounding mucosa edema uplifted ring levee samples, shape such as craters,with white or yellow of necrosis and inflammatory exudate, surroundingmucosa plica disappear, was stripped, ulcer sometimes also with thesurrounding tissue adhesion.The ulcer form of other treatment group rats wasclose to the model group, but the ulcer was shallow and narrow with littleeffusion and sphacelus, no obvious edema and congestion on surroundingmucosa, some still with visible healing scar.2.2The comparison of ucler area and inhibition ratio of uclerCompared with the model group, the ucler area of high-does group,low-does group and control group was obviously deflated which had statisticaldifference (P<0.01or P<0.05). Compared with control group and low-does group,the ucler area of high-does group was obviously deflated which hadstatistical difference (P<0.05). The inhibition ratio of ucler in high-doesgroup was62.7%, which was higher than the27.2%of low-does group and the34.7%of control group.2.3Observation by the light microscopeThe construction of gastric wall was normal, and the endepidermis wereintegrity, glands were line up in order, no obvious infiltration of inflammatorycells in sham operation group rats. In model group rats, the structure of gastricwall was destroyed, and the gastric mucosa appeared necrosis or coloboma,and glands disappeared with disordered arrangement of cells, and mucouslayer with inflammatory cells infiltration. The gastric mucosa of othertreatment groups recovered well, which could be seen that the thickness of thegastric wall increased, and there are new epithelium,and submucosal hadvisible granulation tissue, and had no obvious infiltration of inflammatorycells. The symptoms of high dose group tended to normal, and were similar tothe control group.3The comparison of the expression of EGF and EGFmRNA in each group rats3.1The comparison of the expression of EGF in each group ratsCompared with sham operation group, the expression of EGF in modelgroup were obviously increased in average luminance, and decresed inaverage gray scale, which have statistical significance (P<0.05or P<0.01).After medication, each treatment goup which compared with model group hadobvious increse in average luminance, and decreased in average gray scale,which have statistical significance (P<0.01or P<0.05). Compared in eachtreatment group, high-dose group were obviously incresed in averageluminance, and decreased in average gray scale compared with control group,which have statistical significance (P<0.05), high-dose group were decresedin average gray scale compared with low-dose group, which have statisticalsignificance (P<0.05).3.2The comparison of the expression of EGFmRNA in each group ratsCompared with sham operation group, the expression of EGFmRNA in model group were obviously increased, which have statistical significance (P<0.05). Compared with model operation group, each treatmen group aftermedication all could obviously increase the expression of EGFmRNA, whichhave statistical significance (P<0.01). Compared in each treatment group, theexpression of EGFmRNA was much higher in high-dose group than controlgroup, which have statistical significance (P<0.01), also was much higherthan low-dose group, which have statistical significance (P<0.05).4The comparison of the expression of VEGF and VEGFmRNA in each grouprats4.1The comparison of the expression of VEGF in each group ratsCompared with sham operation group, the expression of VEGF in modelgroup were obviously increased in average luminance, and decresed inaverage gray scale, which have statistical significance (P<0.01). Aftermedication, each treatment goup which compared with model group wereobviously incresed in average luminance, and decreased in average gray scale,which have statistical significance (P<0.01or P<0.05). Compared in eachtreatment group, high-dose group were obvious incresed in average luminance,and decreased in average gray scale compared with control group, which havestatistical significance (P<0.05), high-dose group were decresed in averagegray scale compared with low-dose group, which have statistical significance(P<0.01).4.2The comparison of the expression of VEGFmRNA in each group ratsCompared with sham operation group, the expression t of VEGFmRNAin model group were obviously increased, which have statistical significance(P<0.01). Compared with model operation group, each treatmen group allcould obviously increase the expression of VEGFmRNA, which havestatistical significance (P<0.01). Compared in each treatment group, theexpression of VEGFmRNA was much higher in high-dose group than controlgroup, which have statistical significance (P<0.05), also was much higherthan low-dose group, which have statistical significance (P<0.01).5The comparison of the expression of TGF-β1and TGF-β1mRNA in each group rats5.1The comparison of the expression of TGF-β1in each group ratsCompared with sham operation group, the expression of TGF-β1inmodel group were obviously increased in average luminance, and decresed inaverage gray scale, which have statistical significance (P<0.05or P<0.01).After medication, each treatment goup which compared with model groupwere obviously incresed in average luminance, and decreased in average grayscale, which have statistical significance (P<0.01or P<0.05). Compared ineach treatment group, high-dose group were obviously incresed in averageluminance, and decreased in average gray scale compared with control group,which have statistical significance (P<0.05), high-dose group were decresedin average gray scale compared with low-dose group, which have statisticalsignificance (P<0.05).5.2The comparison of the expression of TGF-β1mRNA in each group ratsCompared with sham operation group, the expression t of TGF-β1mRNAin model group were obviously increased, which have statistical significance(P<0.01). Compared with model operation group, each treatmen group allcould obviously increase the expression of TGF-β1mRNA, which havestatistical significance (P<0.01). Compared in each treatment group, theexpression of TGF-β1mRNA was much higher in high-dose group thancontrol group, which have statistical significance (P<0.01), also was muchhigher than low-dose group, which have statistical significance (P<0.05).Conclusions:1Disperse the depressed liver-energy and invigorate the spleen couldsignificantly improve pathomorphology of gastric tissue in gastric ulcer rats,and improve the ulcer inhibition rate, so as to promote ulcer healing.2Disperse the depressed liver-energy and invigorate the spleen couldimprove the expression of EGF and EGFmRNA, and strengthen the repair ofgastric mucosa, so as to promote ulcer healing.3Disperse the depressed liver-energy and invigorate the spleen couldimprove the expression of VEGF and VEGFmRNA, and promote angiogenesis, so as to improve the quality of ulcer healing.4Disperse the depressed liver-energy and invigorate the spleen couldimprove the expression of TGF-β1and TGF-β1mRNA, and regulate cellgrowth and differentiation, so as to promote ulcer healing.
