Based on the previous study in our group, the purpose of this paper is to evaluate the immuno-modulating effects of peptidoglycan microspheres and their protection effects for mouse against pseudomonas aeruginosa infection, thus providing a theoretical basis for the development and application of immuno-stimulants.Peptidoglycan microspheres were prepared by emulsion-crosslinking method. The drug loading capacity (LC) and encapsulation efficiency (EE) of microspheres were (46.41±0.829)g/100g and (82.80±1.74)%, respectively. Fourier transform infrared spectroscopy (FTIR) analysis showed that peptidoglycan has been encapsulated into chitosan microspheres.Imuunostimulation effects of peptidoglycan microspheres on immune function of mouse were evaluated in this study.200 healthy Kunming mouse were randomly divided into four groups. (1) Control group was given 0.2mL 0.9% asepsis normal saline daily; (2) PG group was given 2mg PG daily; (3) CS group was given 3mg chitosan micropheres daily; (4) PG microspheres group was given 5mg PG microspheres daily. All the groups were gavaged once every 24 hours for 7 days. Eye blood was sampled at 3 d,7 d,14 d and 21 d, respectively. T lymphocyte subgroups in peripheral blood and spleen were detected by flow cytometry. Mouse immunoglobulins (IgA, IgG and IgM) were determined by indirect ELISA. In addition, the clinical behavior, liver and spleen indexes were also monitored. Result: Peptidoglycan microsphere group reveals increased immune organ weight and indexes, as compared with the control groups, which indicates that PG microspheres can promote the growth and development of immune organs, improve the body’s immunity against specific antigens. Peripheral blood and spleen T lymphocyte subgroup detection show the level of CD3+, CD4+,CD3+CD4+ in PG-CS slow-release microspheres group were higher than those in control group, indicating that PG-CS slow-release microspheres can improve the body content of T lymphocyte subsets, and thus has a good role to improve immunity. The mouse IgG levels in PG microspheres group have increased significantly at 14d, and change significantly at 21d (p<0.01). PG microspheres in section 21d,28d were infected with Pseudomonas aeruginosa, the survival rates were 50%,60% and significant difference compared with the control group.The study provides a theoretical basis on physicochemical properties and physiological activity of the oral peptidoglycan, and it is of great significance for development of peptidoglycan microspheres as an immune enhancer.
