Purpose: Compared iodine-rich Chinese herbal compound Decoction with iodine excess on thyroid goiter rat model of TPO expression and thyroid hormone synthesis,clear the treatment of goiter efficacy mechanism.Material and method:SPF level of 52 SD rats were selected. After one week adaptive feeding randomly selected 10 for NC, the remaining 42 were gavaged by PTU,of 1mg/100 g, 14 days after the gavage randomly selected 2 rats of model validation. The 50 rats were randomly divided into 5 groups, MG, CG,HG, IG(each group 10 rats) after model successfully reproduced.NC and MG were administered saline 3ml daily gavage, CG to compound decoction 3ml, HG to be concentrated iodine rich decoction 3ml, IG be potassium iodate solution 3ml,consecutive gavage for 28 days.On the 28 th day,after intervention to intraperitoneal anesthesia, the rats were killed, taken in rat arterial blood, thyroid tissue. Using alkaline ashing arsenic cerium determination of rat serum, the iodine content in thyroid tissue by spectrophotometry, using radioimmunoassay was used to detect the expression of serum T3, T4, TSH, HE staining were observed on thyroid morphology in rats.Determination of TPO expression in rat thyroid using immunofluorescence staining.Conclusion: 1.After gavaged by PTU for 14 days,naked by thyroid follicular significantly unequal in size under light microscope compared with NC,Follicular cavity becomes small, irregular in shape, cavity glial significantly reduced, single layer hyperplasia cubicepithelial cell hypertrophy multicoat, parts of the follicle is formed by the fusion of giant follicle, follicular cavity and interstitial showed a large amount of inflammatory cell infiltration,visible small part of follicular hyperplasia, form a closed cavity free cell group, perifollicular blood vessel rich, prompting goiter model successfully reproduced. 2.The comparison of the relative weight of thyroid, HG and IG the higher the relative weight of thyroid compared with NC, MG, P < 0.05; DG had no statistical difference of thyroid weight compared with NC, P > 0.05. 3.Thyroid morphological comparison:All of CG,HG,IG had been treated.CG returned tonormal; IG still visible follicular cavity and inflammatory cells. 4.Comparison of serum iodine content, CG, HG and IG iodine content increased obviously compared with NC, P < 0.05; CG, HG was lower compared with IG, P < 0.05.The urine iodine content in comparison, CG, HG and IG were significantly increased than NC,P < 0.05; CG and HG was higher compared with IG,P < 0.05; CG urine iodine content was high compared with HG, P < 0.05.Comparison of thyroid iodine content, CG, IG,HG and IG in the thyroid was higher compared with NC, lower contents of MG, P < 0.05;compared with IG, CG, HG was higher, P < 0.05; compared with HG, CG was higher, P < 0.05. 5.Comparison of thyroid function in rats: comparison of TT3, compared with NC,HG was lower, IG was higher content of TT3, P < 0.05; TT4 comparison,content of TT3 compared with NC, IG was higher,MG was lower,P < 0.05;HG, CG and HG TT4 content was lower than IG, P < 0.05. MG was high than NC, P < 0.05;no significant difference between CG and NC,P>0.05.TSH content comparison,compared with NC,MG was higher,P<0.05;NC、CG、HG and IG had no significant different,P>0.05. 6.TPO average optical density is within the thyroid, compared with MG, CG,HG and IG larger, MG smaller, with significant difference, P <0.05. Compared with the IG, CG and a larger group HG, MG smaller, the difference was significant, P <0.05. Compared with HG, CG higher than IG, the difference was significant, P <0.05. 7.Combination of thyroid morphological changes,TPO fluorescence: CG > HG> IG > NC > MG.Conclusion: 1.Compared with an excess of iodine, iodine-rich Chinese herbal compound Decoction can be increased within the body of iodine deficiency thyroid iodine content, but it did not cause iodine hyperthyroidism. 2.iodine-rich Chinese herbal compound Decoction recovery in rats on thyroid morphology and expression of thyroid TPO are superior to excess iodine.
