| Purpose: This study to establish in vitro model of rat glomerular mesangial cell proliferation,and the method of serum pharmacology, observing renal soup flavored jade on rat glomerular mesangial cell proliferation and the influence of MMP- 2 / TIMP- 2, investigate renal soup flavored jade prevention role of glomerulonephritis.Materials and methods: 1. By the method of serum pharmacology, preparation of renal soup flavored jade containing medicine serum. 2. Use angiotensin Ⅱ as a stimulating factor, induced in vitro cultured rat glomerular mesangial cells proliferation. 3. Use determined by MTT method rat glomerular mesangial cell proliferation. 4. Use ELISA method to detect matrix metalloproteinases- 2 and the expression of tissue inhibitor- 2.Results:1.The rat glomerular mesangial cell proliferation:Higher than the blank group, model group OD value,and two groups of more significant differences(P < 0.01); Renal soup flavored jade low, medium and high dose group and the OD value of valsartan group were less than model group, and compared with the model group were significantly different(P < 0.01).2.Soup flavored jade kidney of rat glomerular mesangial cells secretory expression of MMP-2:In Ang Ⅱ stimulation, the expression of MMP- 2 levels significantly decreased, and there was a significant difference compared with the blank group(P < 0.01); Renal soup flavored jade low, medium and high dose group and the expression of MMP- 2 in valsartan group levels were higher than that of model group, and compared with the model group were significantly different(P < 0.01); Renal soup flavored jade low, medium and high dose group compared with valsartan group were significant differences(P < 0.01).3. Soup flavored jade kidney of rat glomerular mesangial cells secrete TIMP- 2 expressionIn Ang Ⅱ stimulation, the expression of TIMP- 2 levels increased significantly, and there was a significant difference compared with the blank group(P < 0.01); Renal soup flavored jade low, medium and high dose group and the expression of TIMP- 2 in valsartan group were lower than that of model group, medium,high dose group and valsartan group were significantly difference compared with model group(P < 0.01); Renal soup flavored jade low dose group compared with valsartan group there were significant differences(P < 0.01), medium dose group compared with valsartan group difference was statistically significant(P < 0.05).Conclusion:1. Renal soup flavored jade has significant inhibition of angiotensin Ⅱ under the stimulus of glomerular mesangial cell proliferation in rats.2. Jade renal soup flavored with significant increase matrix metalloproteinase- 2(MMP- 2) expression of the function.3. The renal soup flavored jade have significantly inhibit MMPS inhibitor- 2(TIMP- 2) expression of the function.4. Renal soup flavored jade through influence the expression of MMP- 2 and TIMP- 2 levels, so as to play the role of prevention and treatment of glomerulonephritis. |
