Background:Ulcerative colitis is a chronic nonspecific colitis with unknown etiology. Most scholars believe that the disease is caused by many factors, genetic、 immune、 environment、infection、.intestinal microbes and other factors involved in the pathogenesis of ulcerative colitis.At present, steroid hormone andsalicylic acid and immune inhibitors were the main medicine in the clinical treatment, but the effect is not satisfactory, and long-term application will have a lot of adverse reactions.China’s traditional Chinese medicine believes that ulcerative colitis is due to spleen deficiency and dampness caused, treated mainly by strengthening the spleen and eliminating dampness, practice of some Chinese herbal medicine in the treatment of ulcerative colitis, made a certain effect. For example Chinese medicine Kangfuxinye in the clinical treatment of ulcerative colitis achieved satisfactory curative effect in recent years, but its mechanism is not clear. In this study, the effect of Kangfuxinye cure inflammatory injury of DSS-induced murine colitis model were studied through analysised the Disease activity index (DAI), Tissue General score and Histolgical score of murine model, the levels of IL-1β、 IL-4、 TNF-α、 INF-Υ、 NF-kB in the colon tissue were also measured.Aims:Evaluate the therapeutical effect of Kangfuxinye and protective mechanism in DSS-induced murine colitis model, Methods:Seventy C57 mices(18-23g) were devided into 6 groups randomly。 Blank group weren’t treated, the remaining group drank 3.5% dextransulfatsodium for 5 days,set up DSS-induced murine colitis model,they were devided into model group、 group of kangfuxinye、 group of higher kangfuxinye、 group of fufanghuangbaiye、 group of 5-ASA。 Model group drank normal water;group of kangfuxinye and group of higher kangfuxinye were devided into 3 groups again;respectively were given intragastric administration, intragastric administration+enemata, enemata;group of fufanghuangbaiye and group of 5-ASA respectively were coloclystered.After 8 days of treatment, all groups were killed。 During this period, we record the Disease activity index (DAI), the Tissular General score and the Histological score.We measured the levels of IL-1β、 IL-4、 TNF-α、 INF-Υ、 NF-kB in the colon tissue.Results:1、 The DAI、 the Tissular General score and the Histological score of DSS-induced murine colitis model were significantly elevated (P<0.05).The expression of inflammatory factor IL-1β、 TNF-α、 INF-Υ、 NF-kB were significantly elevated (P< 0.05).The inflammatory factor IL-4 was decreased significantly (P<0.05).2、 DAI:Group of Kangfuxinye intragastric administration+enemata、group of Kangfuxinye enemata, group of higher Kangfuxinye intragastric administration+enemata 、 group of higher Kangfuxinye enemata, group of FufangHuangbaiye, group of 5-ASA were significantly decreased compared with the model group.Group of Kangfuxinye intragastric administration+enemata、 group of Kangfuxinye enemata were significantly decreased compared with the Kangfuxinye intragastric administration (P<0.01).Group of higher Kangfuxinye intragastric administration+enemata、 group of higher Kangfuxinye enemata were significantly decreased compared with the higher Kangfuxinye intragastric administration (P< 0.05).The Tissular General score:Group of Kangfuxinye enemata were significantly decreased compared with the model group (P<0.01).Group of Kangfuxinye enemata were significantly decreased compared with the model group ((P< 0.05).Group of Kangfuxinye enemata were significantly decreased compared with the Kangfuxinye intragastric administration (P< 0.05). Group of Kangfuxinye enemata is better than Group of Kangfuxinye intragastric administration and group of high Kangfuxinye enemata (P<0.05). Group of Kangfuxinye enemata is better than Group of Fufang Huangbaiye (P<0.05).Compared with group of group of 5-ASA didn’t change. The Histological score:Group of Kangfuxinye enemata、 group of higher Kangfuxinye enemata, group of FufangHuangbaiye, group of 5-ASA were significantly decreased compared with the model group (P<0.05).Group of Kangfuxinye enemata were significantly decreased compared with the Kangfuxinye intragastric administration (P<0.05).Group of higher Kangfuxinye enemata were significantly decreased compared with the higher Kangfuxinye intragastric administration (P<0.05).3、 IFN-y:Group of Kangfuxinye intragastric administration+enemata% group of Kangfuxinye enemata, group of higher Kangfuxinye intragastric administration+enemata 、 group of higher Kangfuxinye enemata, group of FufangHuangbaiye, group of 5-ASA were significantly decreased compared with the model group (P< 0.01).Group of Kangfuxinye enemata were significantly decreased compared with Kangfuxinye intragastric administration (P<0.05).Group of higher Kangfuxinye intragastric enemata were significantly decreased compared with higher Kangfuxinye intragastric administration (P< 0.05).Group of Kangfuxinye intragastric enemata 、 group of Fufanghuangbaiye and group of 5-ASA didn’t change.IL-1β:Results with IFN-Υ.TNF-a:Group of Kangfuxinye intragastric administration +enemata、 group of Kangfuxinye enemata, group of higher Kangfuxinye intragastric administration+enemata、group of higher Kangfuxinye enemata, group of Fufanghuangbaiye, group of 5-ASA were significantly decreased compared with the model group (P< 0.05).Group of Kangfuxinye intragastric administration+enemata、 group of Kangfuxinye enemata ere significantly decreased compared with Kangfuxinye intragastric administration (P<0.05).Group of Kangfuxinye enemata is better than group of FufangHuangbaiye, and compared with group of 5-ASA didn’t change. NF-KB:Group of Kangfuxinye intragastric administration+enemata、group of Kangfuxinye enemata, group of FufangHuangbaiye, group of 5-ASA were significantly decreased compared with the model group (P< 0.05).Group of Kangfuxinye enemata is better than Group of Kangfuxinye intragastric administration (P<0.05).Compared with group of FufangHuangbaiye and group of 5-ASA didn’t change.IL-4: Group of Kangfuxinye enemata and group of higher Kangfuxinye enemata were significantly elevated compared with model group、 group of Kangfuxinye intragastric administration-. group of higher Kangfuxinye intragastric administration、 group of FufangHuangbaiye and group of 5-ASA (P<0.05)Conclusion:1、 Evaluation from the DAI score、 the Tissular General score、 the Histological score,and the levels of the cytokines,Kangfuxinye enemata can relieve the inflammation in colon. Kangfuxinye enemata and 5-ASA enemata has a similar effect. Kangfuxinye enemata is better than Kangfuxinye intragastric administration. Kangfuxinye enemata is better than higher Kangfuxinye enemata. Group of Kangfuxinye enemata is better than Group of Fufang Huangbaiye in the Tissular General score.2、 The Therapeutical effect of Kangfuxinye involves restraining the abnormal inflammation reaction and downing the level of IL-1β、 TNF-α、 INF-Υ、 NF-kB and up-regulating level of IL-4.Thus inhibiting the inflammatory reaction mediated by Thl,the effect of Th2 mediated inflammatory reaction was not obvious.
