Microspheres and nanotechnology for drug delivery have different forms of nanotechnology such as microparticles, liposomes, nanoparticles. These new drug forms can change the pharmacokinetic parameters, and may cross biologic barriers, and has prospect future in many fields. This paper mainly studied the application of nanotechnology in treating bone diseases and vaccine field. There are three parts of experiments.1. Icariin-loaded PLGA nanoparticles:Preparing icariin-loaded PLGA nanoparticles by ultrasification emulsification and studying their physical and chemical properties, realease of icariin, and the effect on human osteoblast cells in vitro. The PLGA nanoparticles had uniform particle size and showed the best property of sustaned realease and biocompatibility. Meanwhile, the icariin nanopaticles had no effect on the growth of MG-63 cells, but promoted the differentiation of osteoblast cells.2. Nanocapsules for bone targeting:A dual hard-template approach coupling with coordination interaction between TA and modified gelatin was exploited to prepare nanocapsules. ZIF-8 was utilized to mediate the formation of nanocapsules shell and lumen. The surface nanocapsules were connected to target molecules. The nanocapsules had uniform particle size and showed the best property of sustaned realease.Meanwhile, they showed good bone affinity.3. OVA-loaded microcapsules:A dual hard-template approach coupling with coordination interaction between TA and Al3+was exploited to prepare miacrocapsules. CaCO3 were utilized to mediate the formation of nanocapsules shell and lumen. Microcapsules prepared by this method were pH responsive. By vitro experiments, OVA-loaded miacrocapsules showed the good biocompatibility.
