Bioreactor is a kind of equipment, which provides environment and culturescells to obtain pharmaceutical protein. Impeller is one important component ofbioreactor and power source for cell culture, which can transfer nutrient. However,impeller will also produce shear force which is harm to cells, an optimized impellerstructure and parameter is a key problem in bioreactor design.According to the feature of Elephant Ear impeller, this paper adopted VOFmode and K-epsilon Mode to simulate two phase mixing field in bioreactor andanalyzed the field of velocity and shear force. Results showed that mixing fluidshape depends on impeller installation method and the vertical installation methodwith single vortex field is better for mass transfer. The comparison betweenElephant Ear impeller and others showed that Elephant Ear has better performanceon shear force and mixing aspects.This paper designed orthogonal experiment to optimize impeller structureparameters, such as impeller width, installation angle and ratio between impellerdiameter and tank inner diameter. We got the optimum size is impeller width120,installation angle45and the ratio1:3. Then an impeller is designed in100Lbioreactor, according to the optimum value in5L. At last, we manufactured5LElephant Ear impeller with optimum size and got better CHO(Chinese HamsterOvary) culture data with it.
