Cell activity is an important part of life forms, biotechnology has different lifefunctions due to the various morphological differences, in order to distinguish thesedifferences and functional morphology studies, sorting cells has important practicalsignificance.With micro-processing technology continues to evolve and mature, whichgreatly promoted the development of microfluidic chips, and microfluidic chips forcell sorting has unique advantages. Miniaturized cell sorting method, not only savesthe test sample, but also greatly shortens the study period. In combination with othermethods of operation may be achieved non-polluting, high throughput, sterile cellselection.By studying microfluidic chip cell sorting methods, we summarized andpresented a novel, non-marking, high-throughput microfluidic chip for sorting cells.With the difference of traditional two-dimensional chip, this study used a double-dimensional microfluidic structure, the structure formed by the intersection cellselection area, combined with laser technology, and microfluidic chip achieve to cellsorting.Using multi-physics numerical simulation software COMSOL Multiphysicssoftware to analysis, simulation. We study the structures of sorting cell microfluidicchannel and high aspect ratio microfluidic chip composition simulated by setting theboundary conditions, initially validated the possibility of sorting cells in microfluidicchip.In the intersection region of the double channel with laser, the experimentalvalidate th cell sorting. By Yeast cell suspension we study the actual cell sortingcontroled the aspect ratio of the microfluidic chip; optional silica and polystyrenesolution instead of the cell solution and experimental study of the efficiency of thecell sorting.Experiments show that cell sorting method using double layers PDMSmicrofluidic chip structure with a combination of laser, is a non-marking,high-throughput cell-based sorting method with high sorting efficiency. Microfluidiccell sorting method combining with laser cell has no pollution, greatly improve cellsurvival.In recent years, with the development of technology, microfluidic cell sorting combining with laser methods developed and gradually matured, the type andefficiency of the filter cell are increased. However, cell-based sorting method onmicrofluidic chips with laser still exist some shortcomings, needing for furtherexploration and research.
