Studies On Taxonomy, Cultivation And Molecular Phylogenetics Of Coccomyces On Broad-Leaved Trees From Hubei And Jiangxi Provinces

Members of Coccomyces which collected from Shennongjia forestry region of Hubei province and the Mount Sanqingshan National Park of Jiangxi Province were detaliedly studied with the most advanced poly-character integrated taxonomy method. And at the same time, specimens of Coccomyces occurring on broad-leaved trees which were deposited in AAUF (the Reference Collection of Forest Fungi of Anhui Agricultural University, China) were classified, identified and complementally studied. Seven species, Leptostromopsis rhododendri, Coccomyces alienus, C. delta, C. huangshanensis, C. hubeinesis, C. mucronatus and C. sinensis were identified. Among the taxa above, L. rhododendri and C. alienus are new fungi and Chinese and English morphological features, Chinese comments, iconographies, hosts and habits were given as well. New geographical distributions and host records were added to the remaining species which were complementally studied. Dichotomous key to these species were complied.At the same time, the author isolated and purified some fungi of Coccomyces which were collected from the Mount Sanqingshan National Park of Jiangxi Province for comparing their cultural characteristics. The results showed that the cultural features of the same strain were not the same on MA, OA and PDA culture media, and different strains of the same Coccomyces species sometimes semblable as well as different at times. Generally, Coccomyces fungi were prone to produce conidiomata on the media, with deficient nutrition (like OA) and some of them could secrete wine red or yellow water-soluble pigment in some conditions.At last, fresh materials of Coccomyces collected from Shennongjia forestry region, the Mount Sanqingshan National Park, Tiantangzhai National Forest Park, Wanfoshan National Forest Park and Huangshan scenic spot were isolated and purified for obtaining pure strains. T, C, A and G content of gene sequences of seventeen strains testing by ourselves and eight strains which were downloaded from GenBank were analysed by the method of MEGA4.1. Then phylogenetic tree of Coccomyces fungi was established by using gene sequence of outgroup Lophodermium pinastri and the twenty five gene sequences aforesaid in software PAUP*4.0b4a. The results displayed that geographical conditions maybe had a big impact on the phylogenetic relationship of intraspecific and intraspecific crossing strains of Coccomyces. And the genetic diversities and otherness of Coccomyces intraspecific strains could be affected by hosts. Morphology and anatomy characteristics of Coccomyces fruitbodies were very useful in the taxonomy on the level of species. As a supplementary mean on the taxonomy and identification of fungi, the molecular biology technology had played an important role in the taxonomy of fungi for its accuracy, convenience and celerity. However, the genetic diversities and otherness of fungi sometimes can lead to deviations of the molecular biology technology. So the identification of fungi still need to be based on the traditional taxonomy, and combining with their ecotope, cultural characteristics, individual auxology and molecular biology, etc. Only then can we get ressonable and scientific achievements.
