This thesis consisits of two sections.Firstly,The taxonomy of Rytismataceae Chevall.on trees from Jiangxi,Sichua,Hainan and Taiwan province were researched in the aspects of micro-morphology,developmental morphology,ecology and geographical distribution according to the taxonomy principles of Darker(1967).The total length of ITS sequence and the A,T,C,G and G+C contents of the studied strains based on the biotechnology,also the relationship of intergenera,interspecies and intraspecies were explored by establishing phylogenetic trees in the second part.Detailed presentation is following:1.The Rytismataceae dry samples which came from four province of the South China and deposited in the Reference Collection of Frest Fungi of Anhui Agricultural University,China(AAUF)were checked and studied mostly based on morphology.Twelve species in five genera of Rytismataceae were identified.Among them,Coccomyces taizhongensis,Lophodermium quadrisporum,L.wuzhishanense and Terriera linderae were new species,and other species,i.e.L.agathidis,L.conigenum,L.petrakii,L.picea,L.pini-excelsae,R.punctatum and R.Salicinum were already known in China.Description,disscusion,habital,line drawings and photographs were provided for the new fungus;and simple description and discussion of morphologic characteristics were given for the other species.Keys were compiled for identification of all species.2.The phylogeny of Rytismataceae from China was studied.The related Genomic DNA were extracted from the tested strains by the method of CATB.The required sequences ITS and 28 S regions were amplified by PCR and sequenced.Phylogenetic trees were established based on the sequences of rDNA-ITS,28 S and the combined sequences by using Leotiia lubrica as outgroup,combing with morphological classification method,the species classification status was derermined.The analysis of phylogenetic tree showed: Species in the same family or genus were usually clustered into a group with strong bootstraps.Ecological environment,geographical postion and the host had effecte on the phylogeny.Some species being placed in different genera,even different family with different morphology could also be clustered together.However,deficiency existing in the research,it's not enough to study the phylogeny of fungus only by the comprehensive analysis based on single or two-gene sequences,more gene sequences need for the deep research.For all this,the traditional morphology method is still the main reasonable basis in fungi classification.The scientific and reasonable results will be achieved combining morphology,phylogeny,genetic physiology and biochemistry and nutritional components with molecular biotechnology of fungi.
