There are many species of long-horned beetles in the natural environment. They are widespreading and damagine. The appraisal method of them is important for prevention. However, it’s unclearly about the specific chromatogram on Cytotaxonomy method for many species. So that, it’s unable identify the characteristics of karyotypes use the characteristics of chromosome. The Cytotaxonomy method is used in this experiment on physiology and biochemistry. The tissues with exuberant differentiation (testis, ovary and midgut) of nine species of Lamiinae, five species of Cerambycinae and two species of Prioninae beetles were chosen for preparing slides of chromosomes under different conditions and used the DPS to analysis the diversity of chromosome among the different species. The results showed that their chromosomal number is 2n=20 and the sex-determining mechanism is Xyp.1. Among the Lamiinae, the chromosome of Olenecamptus cretaceus marginatus, Olenecamptus octopustulatus chinensis, Apriona germari, Psacothea hilaris, Monochamus alternatus, Anoplophora glabripennis and Mesosa myops has the same form. It is composed of 5 pairs of large autosomes,4 pairs of midsize autosomes and 1 pair of medium-size sex-chromosome, and the karyotype formula is 5L+4M+Xyp. The chromosome of Batocera lineolata is composed of 4 pairs of large autosomes,5 pairs of midsize autosomes and 1 pair of minitype sex-chromosome. The karyotype formula is 4L+5M+Xyp. For the Moechotypa diphysis is composed of 6 pairs of large autosomes,3 pairs of midsize autosomes and 1 pair of midsize sex-chromosome. The karyotype formula is 6L+3M+Xyp.2. Among the Cerambycinae, For Trichoferus campestris, Aromia bungii and Trirachys orientalis, their chromosome is all composed of 5 pairs of large autosomes,4 pairs of midsize autosomes and 1 pair of medium-size sex-chromosome, and the karyotype formula is 5L+4M+Xyp. The chromosome of Xysrocera globosa is composed of 6 pairs of large autosomes,3 pairs of midsize autosomes and 1 pair of midsize sex-chromosome. The karyotype formula is 6L+3M+Xyp. The chromosome of Massicus raddei is composed of 4 pairs of large autosomes,5 pairs of midsize autosomes and 1 pair of midsize sex-chromosome. The karyotype formula is 6L+3M+Xyp.3. Among the Prioninae, The chromosome of Dorysthenes fossatus is composed of 3 pairs of large autosomes,6 pairs of midsize autosomes and 1 pair of midsize sex-chromosome. The karyotype formula is 3L+6M+Xyp. The chromosome of Megopis sinica is composed of 4 pairs of large autosomes,5 pairs of midsize autosomes and 1 pair of midsize sex-chromosome. The karyotype formula is 4L+5M+Xyp.The comparative on data handing of nine long-horned beetles of Lamiinae showed that the chromosome of number 3,6,8 and 10 has very significant differences on relative length. The chromosome of number 2,4,5 and 7 has significant differences. Among five long-horned beetles of Cerambycinae, the chromosome of number 1 and 2 has no significant differences on relative length, but greater than the other three species, the number 7 less than the other three species. Between two long-horned beetles of Prioninae, the chromosome of number 1,2,4 and 8 has very significant differences on relative length, the number 9 has significant differences, and the number 5 has no significant differences.
