Acaudina Molpadioide is a kind of sea cucumber, belonging to Molpadidae Family, Holothurioidea Class, Echinodermata Phylum. It is distributed widely among Chinese coastal areas and the resource is considerably abundant. The body wall of sea cucunmber is rich in collagen. And the collagen peptide, the hydrolysate of collagen, has many phys- iologicial functions such as lowering blood pressure, enhancing immunity, improving memory, repairing cartilage and so on, with many applications in food, medical and other fields. At present, the research about collagen peptides from sea cucumbers were mostly focused on the biological activity ,but little on the skincare activity .In this study Acaudina Molpadioide used as raw materials, the extraction of collagen peptides with MW<10 kDa(molecular weight is less than 10000 Dalton) was conducted, during while the skincare activity of different molecular weight range (5 kDa <MW <10 kDa, 2 kDa<MW <5 kDa, 1 kDa <MW <2 kDa, MW <1 kDa) of Acaudina Molpadioide collagen peptides were explored. Then body lotion containing Acaudina Molpadioide collagen peptides was manufactured, and its stability, safety and efficacy were evaluated The main contents in the paper were concluded as follows:The bioenzymatic methods were adopted for the extraction of Acaudina Molpadioide collagen peptides. Firstly, the unwanted protein was removed by sodium chloride solution, through single factor experiment to select the optimum conditions: ten times weight of materials, the concentration of 9% sodium chloride solution to immersion for 24 h. Secondly, four kinds of enzymes including AS.1398 neutral protease, flavor enzyme, compound protease, were used to hydrolyse the body walls of Acaudina Molpadioide and the optimum terms were comformed based on the absorption rate and moisture rate of collagen peptides. The results showed that AS.1398 neutral protease had a better absorption rate and moisture rateof the collagen peptides. On the basis of single factor experiment and orthogonal experiment, the technological parameters for extracting Acaudina Molpadioide collagen peptides were optimized, containing temperature 50℃, enzyme concentration 2400 U/g, enzymolysis time 3 h, and pH 7.5. In terms of these optimized conditions, the absorption rate and moisture rate of the extracting Acaudina Molpadioide collagen peptides with MW<10 kDa, are 6.11% and 81.63% respectively.The skincare activity, containing moisture and absorption, antioxidant activity, and the activity on inhibiting tyrosinase of the Acaudina Molpadioide collagen peptides of different molecular weights were studied. The results of moisture and absorption experiments showed that Acaudina Molpadioide collagen peptides of different molecular weights played well roles in absorbing moisture and holding moisture, with the best effect on collagen peptides that of molecular weight was 1 kDa < MW <2 kDa; The Acaudina Molpadioides collagen peptides of different molecular weights were also used to scavenge (·OH) and (O2-·) to investigate the activity of antioxidantion. The results indicated that the Acaudina Molpadioides collagen peptides of different molecular weights had different scavenging ratios, and the collagen peptides with 1 kDa < MW <2 kDa had the highest scavenging ratio. When the concertration was 20 mg/mL and 22 mg/mL, the scavenging ratio on (·OH) and (O2-·) was 76.77% and 81.76% respectively. Then the biochemical enzymatic methodes were applied to determine the activity of inhibiting tyrosinase by the Acaudina Molpadioides collagen peptides. The results showed that in some concentration, all molecular weight ranges of the Acaudina Molpadioide collagen peptides could inhibit the activity of tyrosinase more or less, with the best effect of molecular weight among1 kDa < MW <2 kDa. In the concentration of 100 mg/mL, the inhibition ratio reached to 89.63%, which is counterpart to the maximum inhibition ratio of arbutin 92.06%.Lastly, the body lotion containing Acaudina Molpadioide collagen peptides were developed, and the safety and efficacy properties were evaluated by vitro and animal expriments. The results demonstrated that the body lotion was stable and safty, with all performances meeting the national light industry standard QB /T1857-2004, and played a great role on preserving moisture.
