Amelanchier alnifolia Nutt.(Saskatoon berry) is a fruit-bearing shrub native to thesouthern Yukon and Northwest Territories, the Canadian prairies and northern plains of theUnitied States (Mazza and Davidson,1992; Mazza,2005). Amelanchier alnifolia Nutt.belongs to the Rosaceae family and the Amelanchier genus. It is an important economic treedistributed in the interrior of North America, and is also one of the fastest growing fruit of thethird era fruit on the international. According to the research reported by a biologicalextraction company, Amelanchier alnifolia Nutt. had the same level content of antioxidantpolyphenols and phytonutrients with blueberry and lingonberry, the polyphenols, procyanidins,anthocyanidins, chlorogenic acid and other antioxidants it contained showed good effects inreducing the incidence of a variety of diseases such as cancer, heart disease, diabetes and forclinical treatment. A period of time in the future, Amelanchier alnifolia Nutt. extracts willhave broad prospect of development and utilization in functional food, healthy nutrition,natural health products and other fields.This paper takes Amelanchier alnifolia Nutt. which has not yet seen in domestic marketas the research object, by extracting, analysising and studing the stability and oxidationresistance of the procyanidins, intends to provide some information for the domestic andforeign scientific workers in studying Amelanchier alnifolia Nutt. The main research resultsare as follows:Using vanillin-hydrochloric acid method for determining the content of Amelanchieralnifolia Nutt. procyanidins. Take the procyanidins content as an index, through the singlefactor and orthogonal experiments the optimum extraction process was investigated:extraction temperature70℃, ethanol concentration, solid-liquid ratio1:15, extraction time90min, extraction times3times,with the extraction yield60.5%.Extracting Amelanchier alnifolia Nutt. procyanidins with the optimum extraction process,then by macroporous resin adsorption and ethyl acetate extraction for purification, gettingAmelanchier alnifolia Nutt. procyanidins with high purity.Then using UV, FT-IR andUPLC-MS to analysis the structure, preliminary determined the basic composition ofAmelanchier alnifolia Nutt. procyanidins is catechin and its isomeride epicatechin.Researching the stability and oxidation resistance of Amelanchier alnifolia Nutt.procyanidins from several aspects, the results showed: the light will reduce the stability ofAmelanchier alnifolia Nutt. procyanidins; temperature and pH can affect the stability of Amelanchier alnifolia Nutt. procyanidins, the stability is better at low temperature and low pHconditions, as the temperature and pH increase, the stability decreases; metal ion Cu2+andFe2+can significantly reduce the stability of Amelanchier alnifolia Nutt. procyanidins; Vc andNaHSO3may play a protective role for the stability of Amelanchier alnifolia Nutt.procyanidins; Amelanchier alnifolia Nutt. procyanidins have good capacity in reducing Fe3+,chelating Fe2+, scavenging DPPH and hydroxyl free radicals, showing good antioxidantactivity, indirectly indicates its broad prospect of development and utilization in anti-agingand health care products.
