The juice and wine processing with pomegranate (Punica granatum) fruit will discarda large number of pomegranate seeds annually in China, which contain procyanidinswith anti-oxidation, anti-aging and other physiological activity. The application ofprocyanidins extracted from pomegranate seed would have broad prospects in theindustry development.‘Dabenzi’,‘Erbenzi’ and ‘Bopicao’, the typical varieties ofHuaiyuan pomegranate were chosen as raw materials in this experiment to analyzeessential ingredients and secondary metabolites. The content of procyanidins inpomegranate seed was determined to provide evidence for the selection ofpomegranate variety suitale for procyanidins extraction. Selected ultrasonic wavepower, pH, solid-liquid ratio and ethanol concentration four factors for responsesurface analysis to get the best process conditions of ultrasonic assisted extraction ofprocyanidins, which impacted procyanidins rate most basised on single-factor test,.Researched on adsorption purification technology of procyanidins in oiarse solutionwith macroporous resin, and selected suitable macroporous resin to study adsorptionand desorption process conditions. Determined inhibition rate of O2-·, clearance rateof·OH and DPPH· inhibition rate of procyanidins got in tests respectively. Orbitalblood was sacrificed after30d feeding study in mice, then determined the mice liverweight, liver SOD and GSH-Px activity and serum MDA content. Result can be gotby tests.(1) Dabenzi, Erbenzi and Bopicao three varieties of pomegranate juice nutritionanalysis shows that Bopicao has the highest total sugar content for184.3mg/mL;Erbenzi has the highest reducing sugar content for110.9mg/mL; Dabenzi has thehighest crude fiber content for0.74mg/mL; Bopicao has the highest VCcontent for0.472mg/100g juice. Oil fatty acids in Erbenzi seeds have the most species, whichwere detected in22species, mainly about palmitic acid, stearic acid, arachidic acid,behenic acid, linoleic acid, oleic acid, linolenic acid, pomegranate acid andalpha-Eleostearic acid. Procyanidins in pomegranate seed of Bopicao, Erbenzi andDabenzi three varieties content is about54.4mg/g,34.4mg/g,33.8mg/g.(2) Response surface optimization of ultrasonic assisted extraction of pomegranateseed procyanidins optimal conditions for solid-liquid ratio1:33, pH5.0, ethanolconcentration of51%, power125W, procyanidins yield up to4.40%under these conditions, relative error between the real value and predictive value was0.23%.(3) Selected the AB-8macroporous resin, initial concentration of crude liquidcolumn was20mg/mL, solution pH4.5, and setted shaker30℃,110r/min oscillationto fully adsorbed. Four times of column volume of30%and50%ethanol was used toelute procyanidins, elution conditions: analytic solution pH4.5,30℃,110r/minconstant temperature oscillation to collect the flowthrough procyanidins purity of85.3%.(4) In the determination of the concentration range, pomegranate seed procyanidinsand VCinhibition rate of O2-·, clearance rate of OH, scavenging rate of DPPH withincreasing concentrations gradually strengthen. The same concentration of VCoxidation resistance is better than pomegranate seed procyanidins. Showed that inmice30days feeding trial, the middle dose group pomegranate seed originalanthocyanins can significantly reduce the serum MDA levels significantly increase therole of the liver tissue SOD and GSH-Px vitality, but high doses of pomegranate seedsoriginal anthocyanins will the livers of mice caused a certain degree of damage.
