The application of aloe has developed into different products such as apparel,household textiles,nonwoven and so on.The aloe finishing of textiles,can produce a moist,soft and comfortable hand feeling,and has moisturizing antibacterial function.The generally recognized active substances in aloe are mainly anthraquinone compounds like aloin and polysaccharide.The active components of products which are obtained through different production extraction processes have great differences,besides,aloin as one of the main anthraquinone substances has strong effect which can bring about diarrhea,if apply in different textiles can have potential harm to consumer products users especially for infant.Currently,the matured assay testing methods of aloe in textiles on how to judge textile products whether contain aloe active substances and how can the amount of content impact human being are insufficient.The main active assay like aloin,emodin and chrysophanol were chosen as main research substances in the paper.The different pre-treatment extraction methods'impact on the extraction amount of target substances were discussed.HPLC/D AD and LC-MS/MS were adopted to analyze the extraction solution.Also proficiency test between laboratories were launched.1,the study on pre-treatment extraction methods of samplesAfter compared with Soxhlet Extraction,water bath operated and the ultrasonic bath,the ultrasonic bath could be used as the most suitable method to extract analyte in textile.Methanol,ethanol,acetonitrile and acetone as extraction solvent were chosen,and the same ultrasonic bath conditions were adopted.Methanol was the best extraction solvent which was indicated through the comparison of analyte results in experiments.The optimum extraction conditions through ultrasonic bath extraction of three analyte were determined as 1:10 of solid to liquid ratio,30min of extraction time,60 ? of extraction temperature,one time ultrasonic extraction of samples was chosen.2,study on assay testing methods of Aloe in Textiles by HPLC/DAD methodIn order to obtain good chromatographic peak shape and less analytical time,discuss chromatographic condition.By analisizing and comparing the methanol and 0.1%phosphoric acid as mobile phase were chosen.Discuss different chromatographic column temperature,the analyze temperature of 30 ? which was reach to room temperature was chosen.358nm was chosen as UV testing wavelength of aloin.256 nm was chosen as testing wavelength of aloe emodin and rhubarb.The methods had great linear relation,the correlation coefficient were above 0.999.The precision of equipment was evaluated through within-day and between-day precision of the standard solvent.The precision of methods was studied by calculating RSD,and stated the method had good repeatability.The study found that the test solution shouldn't be placed under room temperature for long time,and should be measured quickly or the solution was not stable enough when kept in 4 ? refrigerator.3 study on assay testing methods of Aloe in Textiles by LC-MS/MS methodIn order to obtain good chromatographic peak shape and high sensitivity,Liquid chromatography conditions needed optimized.The research found that acetonitrile and 5.75 of pH,10 mmol/L of ammonium acetate were chosen initially as mobile phase.ESI negative ion model was chosen to mass spectrum scanning of analyte.Through analyzing the linear regression data,test limit and quantitative limit,regression curve had good linear relation.The equipment precision was evaluated by repetitive analysis of standard work solution which had different concentration.The accuracy of analysis method was studied through blank standard addition recovery rate and sample standard addition recovery rate.4,comparison of HPLC and LC-MS/MS methodComparing HPLC and LC-MS/MS,the former had better precision and accuracy but higher test limits when testing aloe assay in textiles.LC-MS/MS had high sensitivity,low test limits,better selectivity and wasn't susceptible to interference of sample matrix.Therefore,HPLC was chosen to test when assay of aloe in samples were larger.LC-MS/MS was chosen to test when assay of aloe in samples were smaller or the method had impurities interference.However,the test limit of LC-MS/MS was the price of equipment which was expensive,large scope promotion had difficulties.The two analysis methods in the paper can be applied to aloe assay testing in textiles.While because of the missing relative domestic standards,promotion work of the methods needs to be strengthened.
