The extraction and purification technology of Lithocarpus Polystachyus Red flavonoids were studied experimentally. The effects of extraction and flocculation process on extraction efficiency、loss rate were investigated. Functionalized resins PS-GA and PS-CD were synthesized which had higher selectivities and capacities for dihydrochalcones. The adsorption kinetics、adsorption isotherms and the purification effects of one-step column chromatography were investigated with AB-8and D101as comparison. The adsorption mechanism of dihydrochalcones onto PS-GA and PS-CD were studied preliminary. The main contents can be summarized as follows:(1) The traditional hot water extract had better coincidence with phlorizin in absorbance peaks by comparing with ethanol reflux extraction and ultrasonic extraction as well as better security. And the two-step water extraction efficiency was up to80%. Alcohol sedimentation、PAC and chitosan flocculation were applied in flocculation process. The most optimal flocculant and its consumption were studied with clarity and loss rate of flavonoids as index. AB-8and D101were applied in the one-step purification of Lithocarpus Polystachyus Red flavonoids. The purity of phlorizin were both lower than55%.(2) Two adsorbents functionalized with glucosamine (GA) and (3-cyclodextrin (β-CD) were synthesized and characterized by Fourier transform infrared (FTIR) spectra and elemental analysis. Static adsorption, adsorption kinetics and adsorption isotherms of PS-GA and PS-CD were studied experimentally. Adsorption kinetics of PS-GA and PS-CD had better simulation result with the Expassoc diffusion model and adsorption isotherms of them fit the Freundlich model better. After one-step purification of PS-CD column, the purity of phlorizin purity was up to79.48%, higher than the66.56%of PS-GA column. These two modified resins showed better purification effect of Lithocarpus Polystachyus Red flavonoids(3) In order to explore the adsorption mechanism of PS-GA and PS-CD, the FTIR of the adsorbent PS-GA before and after adsorption of the Lithocarpus polystachyus Rehd leaves extract were detected. The slightly blue shifted after adsorption of NH bond illustrated that the hydrogen bonds adsorption mechanism. The special cavity structure of β-CD grants PS-CD to form inclusion complexes with suitable guest flavonoids molecules. P-CD inclusion complexation might exist in the PS-CD adsorption progress besides hydrogen-bonding interaction, which all contribute to the high selectivity. It is because the strong adsorption that the regeneration ability is poor. However, the resin exhibited good reproduction performance which is important for large-scale production.
