Lithocarpus polystachyus(wall.)Rehd is an evergreen plant of the Fagaceae family Lithocarpus,also known as sweet tea,polystachyus(wall.)Rehd,etc.Its tender leaves taste sweet and refreshing.It is a traditional Chinese herbal medicine and has been used in traditional Chinese folk medicine for a long time.Studies have found that Lithocarpus polystachyus mainly contains flavonoids,triterpenoids,polyphenols and other ingredients,which have a variety of pharmacological effects.The main sweet component is phlorizin in flavonoids,which has many important biological activities such as hypoglycemic,anti-oxidation and anti-cancer,and is widely used in food additives and medical and health care industries.Black tea,a fully fermented tea,has a strong fusion and can be blended with a variety of raw materials to form a product with a more rich aroma and a more unique taste,which can better attract the attention of consumers.Market space.Lithocarpus polystachyus is used as a natural sweetener,its phloridin and other ingredients can be added to black tea beverages with the function of inhibiting the increase in blood sugar,avoiding the use of artificial sweeteners such as saccharin,aspartame,and sodium cyclamate.used,make the product have a certain health effect,this research aims to develop a compound health drink for diabetic patients.The main results are as follows:1 Liquid chromatography-time-of-flight mass spectrometry to determine the processing method of Lithocarpus polystachyus(wall.)RehdUsing liquid chromatography-time-of-flight mass spectrometry to analyze the active ingredients of steam-fixation and roller fixation Lithocarpus polystachyus.The results showed that 65 active ingredients were identified in the steam-fixation samples,and 64 active ingredients were identified in the roller fixation samples,including amino acids,vitamins,polyphenols,flavonoids,alkaloids,fatty acids,and two types of fixation.The composition of the active ingredients is similar under the method.Analysis of its content found that the higher content of the two types of curing methods were flavonoids,but the content(8.06%)in steam fixation samples was higher than that of roller fixation(6.68%).The main sweet component phlorizin was extracted and compared with the main source of natural phlorizin-apple.The results showed that steam fixation(32.60±2.26mg/g)and roller fixation(31.78±1.38mg/g)The content of phlorizin was significantly higher than that of apple peel(0.34±0.03mg/g)and apple pulp(0.04±0.01mg/g).Therefore,in the follow-up experiments,steam fixation Lithocarpus polystachyus was used as the sweetener of the compound beverage.2 Determination of the best formula of Lithocarpus polystachyus(wall.)Rehd black tea compound beverageIn this study,two schemes were used to study the formula of Lithocarpus polystachyus black tea compound beverage.The first scheme was to directly mix Lithocarpus polystachyus powder and black tea powder,and add sour and stabilizers for flavoring to make Lithocarpus polystachyus Blending black tea;the second option is to add Lithocarpus polystachyus powder to the withered tea leaves,knead and ferment to make black tea.Using single factor and orthogonal experiments,using sensory evaluation scores as indicators,it was determined that the best formula for black tea blended with Lithocarpus polystachyus is 2:1 ratio of black tea and Lithocarpus polystachyus,and the addition amount of citric acid is 2%,the?-cyclodextrin addition amount is 0.08%.The best formula for the fermented black tea of Lithocarpus polystachyus is that the ratio of leaves to Lithocarpus polystachyus powder after withering is 9:1(m:m),and the fermentation time is 6h.Compared with the Lithocarpus polystachyus blended black tea of Scheme 1,the antioxidant tea polyphenol content in the Lithocarpus polystachyus fermented black tea of Scheme 2 is higher;the content of caffeine with excitatory nerve effect is lower,which is more suitable for the elderly and children;There is no significant difference in the content of amino acids and soluble sugars.The content of flavonoids and phlorizin is low,but the content of phlorizin is higher than that of apple pulp(0.042±0.014mg/g)and apple peel(0.338±0.029mg/g),The comprehensive evaluation determined that the best formula of Lithocarpus polystachyus black tea compound beverage is that the ratio of withered leaves to Lithocarpus polystachyus is9:1,and the fermentation time is 6h.3 Functional Determination of Lithocarpus polystachyus Black TeaThe in vitro hypoglycemic activity of the optimally formulated compound beverage was tested,and the results showed that its inhibitory rate on the?-glucosidase that decomposes compound sugars into glucose was 76.96%±0.48%,which was slightly lower than commonly used hypoglycemic drugs Acarbose(80.23%±0.17%),higher than ordinary black tea(23.52%±0.74%);the inhibitory rate of?-amylase that decomposes starch and other macromolecules into disaccharides is 62.78%±3.58%,which is low The positive control acarbose(79.10%±2.89%)is higher than that of ordinary black tea(20.39%±0.46),indicating that the optimal formula of the compound beverage has a certain hypoglycemic effect,and the addition of Lithocarpus polystachyus can improve the effect of black tea Inhibition rate of?-glucosidase and?-amylase.The free radical scavenging rate of the compound beverage of the best formula was determined.For ABTS free radical scavenging rate:the scavenging rate of Lithocarpus polystachyus black tea was 96.68±1.57%,which was compared with the positive control ascorbic acid(Vc)(97.22%±1.69%)There is no significant difference.For DPPH free radical scavenging rate:the scavenging rate of Lithocarpus polystachyus black tea is 72.77%±2.66%,and the effect is 81%of the positive control Vc(89.59%±1.01%).The measurement results of hydroxyl radical(OH)scavenging rate show that the scavenging rate increases with the increase of sample amount.When the sample amount is 0.1-0.2m L,the scavenging rate of ordinary black tea is higher than that of Lithocarpus polystachyus black tea;When it is larger than 0.5m L,the scavenging rate of Lithocarpus polystachyus black tea is higher than that of ordinary black tea,indicating that Lithocarpus polystachyus black tea has a higher ability to scavenge hydroxyl free radicals after reaching a certain concentration.The measurement results of superoxide anion(O2-)clearance rate showed that the clearance rate of Lithocarpus polystachyus black tea was higher than that of ordinary black tea,and the clearance rate increased with the increase of sample amount,indicating that the black tea added Lithocarpus polystachyus increased The superoxide anion scavenging ability of black tea.The results of the cancer suppression test of the optimal formula of the compound beverage show that Lithocarpus polystachyus black tea has a higher effect of inhibiting cervical cancer cells than ordinary black tea,and with the increase of the sample concentration,the more obvious the effect of inhibiting cell proliferation;For small cell lung cancer A549 cells,when the sample concentration is 100?g/g,the cell viability is most inhibited,and the inhibitory effect of Lithocarpus polystachyus black tea is higher than that of ordinary black tea,and then with the increase of the sample amount,the cell viability is not significant The difference indicates that Lithocarpus polystachyus black tea has a better inhibitory effect on non-small cell lung cancer A549 cells than ordinary black tea.It shows that adding Lithocarpus polystachyus can improve the anti-cancer effect of ordinary black tea.
