Studies On Extraction And Purification Of Phlorizin In Lithocarpus Polysachyus Rehd

Phlorizin is widely used in food and drugs as healthy sweeteners In current study, phlorizin is extracted from Lithocarpus polysachyus rehd by the method of water extraction under microwave. the influnce brought by several factors, including preliminary extraction time,microwave reaction time,liquid-solid ratio, microwave power and extraction times, the yield of phlorizin is discussed in this paper. In order to calculate the best condition of extraction, the methodology of Box-Benhnken Response surface design is applicated. Accordingly, the best condition of water extraction:when preliminary extraction time is2times,microwave reaction time is6.0min; microwave reaction time is5group;liquid-solid ratio is18; microwave power is533w,extraction times is2; we calculated the the yield of phlorizin was3.09%.A new type of natural polymer flocculant "chitosan" was used to improve the purity of phlorizin instead of the traditional flocculant "polyaluminium chloride". The technological parameters were investigated in term of flocculant dosage, pH, flocculation time and temperature.In order to calculate the best condition of extraction,the methodology of Box-Benhnken Response surface design is applicated. Accordingly, the best condition of purity:when the chitosan flocculant is used at0.54mL/g (dried leaves)(equal to0.63g/L water extract),the pH of the water extract is6.17, the period of the flocculation is72.45min, the temperature of the water is25℃, resulting the transparency "T620nm" can reach97.05%andthe purity of phlorizin can reach21.93%.Finally, phlorizin is further pured by ADS-7macroporous resin. Subsequently, the influnce brought by several factors, including adsorption kinetics,adsorption isotherm,leak curve,eluent concentration and the dosage of eluent,the purity of phlorizin is discussed in this paper.then the following conditions are concluded:the adsorption time is 10h, he best sample concentration is1.99mg/ml,the volume of the sample is3BV, the best eluent is70%ethanol, dosage of eluent is2BV. Phlorizin is purified from21.93%to58.56%.
