As an important synthetic material for drug compounds,(-)shikimic acid, hasbeen widely used in chemical, food, medicine and other fields. Therefore, The studyon (-)shikimic acid resources and its extraction, separation and purification hasimportant significance. In this paper, after culture, fermentation and verification, threestrains which can produce shikimic acid were screened from more than ten soilsamples, the yield of(-)shikimic acid of strains from three soil samples was0.174g/L,0.131g/L,0.087g/L. So they can be used for the development and application ofsubsequent fermentation. The desired products in pure form were obtained byrecrystallization method, IR, MS,1H NMR and Elemental analysis were used tocharacterize the products.In this paper, fermentation conditions of the strains that produced (-)shikimicacid and were screened from soil samples were optimized. the best fermentationconditions were that: the initial pH value of fermentation liquid was7.0, inoculumswas5%, fermentation liquid volume was100mL, fermentation time was91h, underthe condition, the content of shikimic acid is0.946g/L.In this paper, we studied the analysis method of shikimic acid: the content of(-)shikimic acid in fermentation liquid was determined by Ultraviolet–visiblespectroscopy, the detection wavelength was245nm. In the concentration range of0-0.3mg/mL, there was a good linear relationship between the absorbance and thecontent of (-)shikimic acid, the regression equation was A=3.0844C-0.0048,R2=0.9999, the average recovery was100.04%, RSD was0.32%, the accuracy is0.10%. Therefore, this content determination of shikimic acid was simple, rapid andhigh accuracy.(-)Shikimic acid in fermentation liquid was separated and Purificated by717anion exchange resin, and the adsorption characteristics of717anion exchange resinto (-)shikimic acid were studied by methods of static and dynamic adsorption. Theadsorption isotherms among study range of temperature were determined by static adsorption, and the outflow curves were determined by dynamic adsorption, thepenetration curve equation was regressed by model and the all correlation coefficientsof the equation were above0.97. The best adsorption conditions were that: initialconcentration of liquid was0.752g/L, the rate of feed flow was1ml/min, resincolumn (H, D) was40X25mm, temperature was24℃, breakthrough curve equationwasC0.7521exp[(t1.252.64460.6209)]In this paper, The desorption process was studied, the results show that differentconcentrations of ethanol eluent had great influence on the desorption effect. Butconsidering the subsequent process feasibility and economical efficiency of theproduct, the best elution conditions was that: ethanol concentration was95%, theelution rate was1.4ml/min, temperature was room temperature.
