In order to provide basis theoretical for extraction of shikimic acid in the industrial. This topic with fresh pine needles as raw material, the conditions of the extraction parameters of response surface methodology optimization under ultrasonic assisted on shikimate from pine needles, and the separation and purification to ion exchange resin, and optimize their separation and purification process the shikimate the anti-platelet aggregation of features research. The main results are as follows:1. ln order to optimize the extraction parameters of shikimic acid from the pine needles, to shikimic acid as the inspection index, single factor test to determine the solid to liquid ratio of l:30(g/mL); Auxiliary conditions for100W ultrasonic treatment15min;Process variables such as the concentration of ethanol in extraction agent, extraction temperature and extraction time were optimized using response surface methodology on the basis of one-factor-at-a-time experiments. The results indicated that the optimal extraction conditions were22%ethanol as extracting solvent, material/liquid ratio of l:30(g/mL), the ultrasonic processing15min of100W of auxiliary condition, extraction temperature of69℃, extraction time of28min, resulting in an extraction yield of5.0783%.The established regression model describing the extraction yield of shikimic acid from pine needles as a function of three extraction parameters was highly significant(R2r=0.9897). The predictive and experimental results were found to be in good agreement.2. The adsorption and desorption rates of4types of adsorption resins towards shikimic acid from the Pine Needles were investigated. Among the studied ion exchange resin,331anion exchange resin was the most suitable to purify crude shikimic acid from the Pine Needles. When4BV of1.5(mg/mL) sample was flowed through331anion exchange resin column at a speed of2(mL/min), the adsorption of the331anion exchange resin reached dynamic equilibrium. The adsorbed shikimic acid could be completely eluted with8BV of2%sodium hydroxide solution at4(mL/min). After the purification, the purity of shikimic acid was increased from25%to90%.3. Using the method of in vitro drug investigated the pinus massoniana pine needle extract shikimic acid to adenosine diphosphate (ADP) and collagen induced in vitro platelet cond- ensed influence, the test results show that the pinus massoniana pine needle extract shikimic acid can be a very good inhibition adenosine diphosphate (ADP) and collagen induced in vitro platelet clumping, and its dose and inhibition effect on certain positive correlation fun-ction. Proof of pinus massoniana pine needle extract shikimic acid can inhibit the in vitro induced platelet clumping, has the effect of anti-blood stasis in vitro and has certain anti-thrombus function.
