Polysaccharide is one of the important active ingredients of Cyclocarya paliurusleaves. This study was conducted to investigate the antihyperlipidemic and antioxidanteffect of Cyclocarya paliurus (Batal.) Iljinskaja polysaccharide(CPP) in high-fatemulsion-induced hyperlipidaemic rats. We examined the effects of (CPP) on weight gain,serum levels of the total cholesterol (TC), triglyceride (TG), low density lipoprotein(LDL-C), high density lipoprotein (HDL-C) and blood antioxidant enzymes. Theexpressions of HMG-CoA, FAS, ATGL, PPARα, Glut-4were investigated by Q-PCR toexplore the effect of CPP in liver and white adipose tissue of hyperlipidemic rats. Theprotein expression of related genes were performed by western-blotting. The resultsshowed as follows:(1) During the animal experiment, Cyclocarya paliurus (Batal.) Iljinskajapolysaccharide showed significant hypolipemic effect. Serum TC and TG levelswere significantly lowered by CPP compared to the model group(P<0.01) and inmost cases were indistinguishable from rats fed normal chow, basal diet(normalcontrol group). Conversely, levels of high density lipoprotein-cholesterol (HDL-C)were elevated in the groups given daily doses of CPP(P<0.01), compared to thehyperlipidemic model group.(2) Activities of total superoxide dismutase (T-SOD), glutathione peroxidase(GSH-Px) and catalase (CAT) in serum were significantly elevated in CPP-treatedgroups while levels of malondiadehyde (a byproduct of lipid peroxidation)decreased(P<0.01or P<0.05).(3) CPP could significantly inhibit the body weight gain in rats with hyperlipidemia(P<0.01) and significantly reduce the aliphatic index (P <0.05).(4) Cyclocarya paliurus (Batal.) Iljinskaja polysaccharide could significantlyenhance the mRNA expression of ATGL, Glut-4, PPARα(P<0.01or P<0.05) andrestrain the mRNA expression of FAS (P<0.01) and HMG-CoA in liver and (or) inwhite adipose tissue of hyperlipidemic rats.(5) CPP could significantly enhance the protein expression of ATGL, Glut-4, PPARαand restrain the protein expression of FAS, HMG-CoA in liver of hyperlipidemicrats (P<0.01or P<0.05). Cyclocarya paliurus (Batal.) Iljinskaja polysaccharide could also affect the protein expression of FAS, ATGL, HMG-CoA, PPARα,Glut-4in other organs of hyperlipidemic rats.
