Cyclocarya Paliurus(Batal.)Ijinskaja,a Chinese native plant,also known as cash cow,Qingqianli,sweet tea tree,is the sole species in its genus which belongs to the genus Cyclocarya Iljinskaja(Juglangdaceae)and is located at an altitude of 600 ~ 1500 metres in the mountainous regions in Hubei,Hunan,Jiangxi,Guangxi and Guizhou,etc.The leaves of Cyclocarya Paliurus(Batal.)Ijinskaja are rich in polysaccharides,flavonoids,saponins,and some other secondary metabolites.Previous studies have shown that Cyclocarya Paliurus(Batal.)Ijinskaja polysaccharide(CPP)has remarkable hypoglycemic activity.Selenium is essential trace elements,which has some important biological activities,such as antioxidant,immunoregulation,etc.In this paper,we obtained Cyclocarya Paliurus(Batal.)Ijinskaja leaves which were rich in selenium by spraying organic selenium fertilizer artificially.Then,selenium polysaccharide from Cyclocarya Paliurus(Batal)Ijinskaja(Se-CPP)was isolated.Finally,the differences of hypoglycemic,antioxidant and immunoregulation effects between Se-CPP and CPP were compared in diabetic model mice,and the potential mechanisms of hypoglycemic activity of Se-CPP were also discussed.The main research works in this paper are as follows.(1)The Cyclocarya Paliurus(Batal.)Ijinskaja leaves were spraied organic selenium fertilizer artificially.After 20 days,the leaves containing selenium and normal levers were collected and dried fully.Then Se-CPP and CPP were isolated by hot water extraction and ethanol precipitation.The selenium content of levers,Se-CPP and CPP were measured by hydride generation-atomic fluorescence spectrometry.The results showed that the selenium content of Cyclocarya Paliurus(Batal.)Ijinskaja leaves containing selenium was 4.38 mg/kg,which was more than the national standard of Se-enriched tea(0.25 ~ 4.00 mg/kg).Finally,anthrone-sulfuric acid method was used to determine the contents of polysaccharide in the extract.The contents of polysaccharide in Se-CPP and CPP were 39.5% and 37.3%,respectively.(2)The diabetic mice induced by giving high fat diet and the intraperitoneal injection of streptozotocin were fed with different doses of Se-CPP,CPP,CPP+sodium selenite,CPP+selenium yeast and xiaoke pill.After 42 days of administration,the levels of oral glucose tolerance,serum lipid,serum antioxidant enzyme activities and immune activities of diabetic mice were measured.The results showed that the appropriate dose of Se-CPP could significantly reduce postprandial blood glucose,serum total cholesterol in diabetic mice,increase the activities of superoxide dismutase and glutathione peroxidase in diabetic mice,and enhance the immunity of mice.(3)Acarbose was uesd as a positive control drug.The inhibitory rates of Se-CPP,CPP,CPP+sodium selenite and CPP+selenium yeast to ?-amylase and ?-glucosidase were determined successfully.The results showed that the order of inhibition activity of each inhibitor to ?-amylase was: acarbose > Se-CPP > CPP+sodium selenite > CPP+selenium yeast > CPP,and the order of inhibition activity of each inhibitor to ?-glucosidase was: Se-CPP > CPP+sodium selenite > CPP > CPP+selenium yeast > acarbose.(4)Metformin hydrochloride was used as a positive control drug.The effects of Se-CPP on cell proliferation and glucose consumption of HepG2 cells were measured.Insulin resistance HepG2 cell model was induced by high concentration glucose and insulin.Then the glucose consumption of IR-HepG2 cells was determined.The results showed that Se-CPP had no significant influence on the proliferation of HepG2 cells,and could significantly increase the glucose consumption,improve insulin resistance of HepG2 cells,and the effects were better than CPP,CPP+sodium selenite and CPP+selenium yeast.At present,there are no detailed reports about the active constituents of Se-enriched Cyclocarya Paliurus(Batal.)Ijinskaja in the world.The results of this paper might provide important experimental evidence for the research and development in Se-enriched Cyclocarya Paliurus(Batal.)Ijinskaja.
