Liubao Tea belongs to one of famous black tea in China, which has a long history of productionand a variety of biological activity. Processing technology is the primary research, while studies onchemical composition is limited. In this paper, the active chemical composition of Liubao Tea andthe purification process of Theabrowns extraction were studied. The main experiments contents areas follows:1.ActivityAnalysisPetroleum ether, ethyl acetate and n-butanol were used to extract the95%ethanol extractedliquid from Liubao Tea, then the four polarity extracted were studied on the antioxidant, antibacteri-al and anticancer experiments. DPPH radical, hydroxyl radical and superoxide anion radical weretexted in the antioxidant experiments. The antioxidant activity was determined by UV absorbancevalue, with the results datas, ethyl acetate and n-butanol pHase have excellent antioxidant capacity,especially clearance value of DPPH radical is the best. Gram-negative Pseudomonas aeruginosa,Escherichia coli and Gram-positive StapHylococcus aureus, Candida albicans were selected todetect the antibacterial activity about the four pHase with the minimum bacterial concentration. Theinhibition effect on Escherichia coli is better than other pHase. Then human gastric cancer cells(MGC-7901), human cervical cancer cells (Hela), Human liver cancer cells (7404) were selected todetermine the anticancer activity about the four pHase. By measuring IC50values, which showgreatly difference with what were expected, the four phase have small inhibition effect to the threecells.2. Chemical componentsAnalysisThe Petroleum ether and ethyl acetate pHase were tested by GC-MS to determine the volatilecomponents. Then Petroleum ether, ethyl acetate and n-butanol were separated to analysis thecompounds. With the gel column chromatograpHy, nine compounds were obtained, five compoundswere identified by NMR,IR,MS. The five compounds were palmitic acid, Gallic acid, Gallicin,caffenine, α-spinasterol.3. Process of extraction and PurificationTheabrownine was extracted with cellulose auxiliary method, the effects of the enzymedose, enzymatic hydrolysis temperature, enzymatic hydrolysis time and the pH of extraction wereinvestigated by orthogonal test. The best extraction rate with cellulose was24.503%.Macroporous resin was selected to purify Theabrownines, absorption and desorption ratio ofD900, DM-301, ADS-17, S-8, D101to TBs were analyzed with static adsorption. D101wasselected to purify TBs. The absorption ratio is43.65%and desorption ratiois67.09%. The concentration of the TBs, the eluent, the elution rate, pH value were determined in the experiment.Acetone-water as the elution, sepHadex as the packing of the gel column chromatograpHy todo fractionation experiment on TBs, six fractions were obtained according to the difference of thepigments ribbon. Every fraction has similarity with the analyzed of the datas of UV-VIS ang IR.Datas show the composition and the structure of the TBs were complexity.
