Rosmarinus officinalis extract is an efficient antioxidant, which contains many kinds of chemical composition with physiological activation. Based on the free radicals theory of aging, current researches have revealed potential antiaging effect of matter with antioxidant properties. In this paper, the quantitative analysis of major active components and the antiaging ability of Rosmarinus officinalis extract were studied.Firstly, major active components had been quantitated by High performance liquid chromatography(HPLC).The column was ODS column (250mm×4.6mm,5μm); peak spectra was280nm. A binary H2O-0.1%H3PO4(solvent A) and acetonitrile (solvent B) gradients was used for elution. It was showed that three compouds was rosmarinic acid, caffeic acid, carnosic acid.Secondly, longevity test and biochemical analysis methods was used to discuss the effect of Rosmarinus officinalis extract on the lifespan and antioxidant system of Drosophila. Every test groups prolonged the lifespan of Drosophila, with the increase of Rosmarinus officinalis extract, aging was delayed in a dose-response relationship.3mg/mL test group could significantly increase the activities of superoxide dismutase (SOD) and catalase (CAT) and decrease the content of maleic diadehyde (MDA).Thirdly, longevity test and biochemical analysis methods was used to discuss the protected effect and the mechanism of Rosmarinus officinalis extract about the damage of lipid per-oxidation result by used10%lard in fruit flies. The present study foud that dietary fat accelerates the aging process and causes a greater mortality by accumulating lipid hydroperoxide in fruit flies. The mean lifespan and maximum lifespan of fruit flies fed by10%lard diet containing3mg/mL Rosmarinus officinalis extract are higher than the groups fed by10%lard, but lower than the control group;3mg/mL test group increased SOD and CAT activity, decreased the content of MDA. The damage of the lipid per-oxidation result by high fat can be resisted in a certain degree by some concentrations of Rosmarinus officinalis extract.Finally, hydrogen peroxide treatment and paraquat treatment was used to exmine the resistance of flies agaist oxidantive stress. The study revealed that supplementation of3mg/mL Rosmarinus officinalis extract prolonged the survival time of fruit flies in the hydrogen peroxide treatment, a similar trend was observed in the paraquat treatment.The results of experiments showed that hawthorn extract possess strong antioxidant activity and anti-aging activity.
