Amygdalus pedunculata Pall.is a deciduous shrub with perennial rootstock,which belongs to subgen Amydalus, Amygdalus, Prunideae, Rosaceae. It is a sun plant and fine tree species with strong stress resistance, such as drount-resistant capacity,cold-resistant capacity, wind erosion-resistant capacity, which can be used to protect sand from wind, conserve water and soil widely in northwestern China. It has high ornamental value, espscially its beautiful flowers, and high nutritional value, which can be widely used to many products. In this essay, the comparasion of the extraction ratio after petroleum degteasing of five provenances was studied. The ultrasonic extraction process of polyphenol from Amygdalus pedunculata seed and the antioxidant activity of Amygdalus pedunculata seed from five provenances was studied.The influence of temperature, time, ethanol volume fraction, times, and ratio of liquid and material on extraction yield of polyphenols were discussed through orthogonal method based on single factor. And the antioxidant activity of Amygdalus pedunculata seed from five provenances was measured with DPPH, ABTS and FRAP methods. The results showed that:(1)The extraction ratio after petroleum degteasing of samples of all five provenances was65.32±1.31%. The provenances which samples had different extraction ratios after petroleum degteasing were as follows:Yuyang, Shaanxi> Wushenqi, Inner Mongolia> Shenmu, Shaanxi> Guyang, Inner Mongolia>Fengning, Hebei. The extraction ratios after petroleum degteasing of samples of Yuyang and Guyang, Fengning had the significant difference(P<0.05), but the extraction ratios of samples of the other provenances didn’t have the significant difference.(2) The optimal conditions of the ultrasonic extraction process of polyphenol from Amygdalus pedunculata seed were obtained as follows:20%ethanol with lg:20mL ratio of Amygdalus pedunculata to solvent, extracting60min (three times with twenty minutes each time)at40℃, the extraction of polyphenol was2.102±0.061mg·g’1.In the orthogonal method, The factors influencing this extraction efficiency were as follows:ethanol volume fraction> ratio of liquid and material> temperature> time.(3)It showed the different radical-scavenging ability in the samples of five different provenances. The IC50value and VCEAC value of the sample from Shenmu, Shaanxi were 199.90±14.07μg·mL-1and139.65±1.43μmol·g-1, which showed stronger radical-scavenging ability than the samples from the other four provenances(P<0.05). The IC50values and VCEAC values of the sample from Yuyang, Shaanxi and Wushenqi, Inner Mongolia were388.66±9.36μg·mL-1,127.78±3.30μmol·g-1and394.31±8.96μg·mL-1,132.43±1.78μmol·g-1, which showed strong radical-scavenging ability. The IC50values and VCEAC values of the sample from Fengning, Hebei were640.87±24.45μg·mL-1and55.68±3.52μmol·g-1, closed with the sample from Guyang, Inner Mongolia, which both showed weak radical-scavenging ability. It also showed the different reducing power in the samples of five different provenances. The FRAP value of the sample from Wushenqi, Inner Mongolia was112.49±1.55μmol·g-1, which showed the stronger reducing power than the samples from the other four provenances(P<0.05). The FRAP value of the sample from Shenmu, Shaanxi was112.49±1.55μmol·g-1, which also showed strong reducing power. The FRAP value of the sample from Fengning, Hebei was45.56±1.82μmol·g-1, also closed with the sample from Guyang, Inner Mongolia, which both showed weak reducing power. All things considered, the sample from Shenmu, Yuyang, Wushenqi showed the better antionxidant capacity than the other two provenances.
