In the field of drug research, more than one-third of the clinical drug is from the nature of thenatural products. Based on the structure of the active compounds of natural products in nature,Organic chemists are constantly exploring new ways to combine and restructure hydrocarbonmolecules to make new pharmaceuticals with higher activity and low toxicity, and make themwidely used in clinical. This is one of the important ideas in the development of the new drugs.New structural, curative effect and low toxic compounds were got from natural products, whichcould be used for drug research. This dissertation was consisted of two parts:1. The separation, purification, structure conformation of chemical constituents ofCaesalpinia Cucullata Roxb.Caesalpinia Cucullata Roxb. was obtained from Bozhou Guo Shao Tang Tech Co., Ltd.Company of Anhui, powdered and extracted with95%ethanol for three times. Then the resultingmixture, which was obtained via vacuum distillation unit, was diluted with water, and extractedwith equal volume of acetic ether for three times. The resulting mixture, which was obtained viavacuum distillation from the acetic ether layer, was isolated by repeated column chromatography(CC) on sillca gel, RP-18, and Sephadex LH-20, as well as by preparative thin layerchromatography (PTLC) and recrystallization. From Brucea javanica (L) Merr.,15compoundswere isolated. The structures of these compounds were elucidated by means of spectroscopicmethods including1D-NMR (1H-NMR,13C-NMR) and2D-NMR (Noesy), and11compoundswere identified as follows by spectral analysis:1: pimpinellin2: scoparone3: isopimpinellin 4: umbelliferone5: β-Stigmasterol6: artanin7: psoraderm8:2H-1-Benzopyran-2-0ne9:6-Methoxy-7-prenyloxycoumarin10:5,7,8-Trimethoxycoumarin11:6-Benzofuranacrylic acidAmong them6,7,8,9,10,11were isolated from this plant for the first time. pimpinellin inCaesalpinia Cucullata Roxb which grow in shanxi province is the main component of CaesalpiniaCucullata Roxb. It is about0.18%. We can use the content of the pimpinellin to identify theCaesalpinia Cucullata Roxb. This kind of Zanthoxylum dimorphophyllum contains a lot ofcoumarins. But it just contains few alkaloids compounds. People should pay attention to useShaanxi made Zanthoxylum dimorphophyllum as traditional Chinese medicine. It is different withthe sun-backed Chinese prickly ash which grow in other province. Especial low lipid patientsshould pay attention to this point. This study on Caesalpinia Cucullata Roxb. provides a basis forthe use of traditional Chinese medicine.2. The separation, purification, structure conformation of chemical constituents of Pipernigrum L.Piper nigrum L. was obtained from Bozhou Guo Shao Tang Tech Co., Ltd. Company ofAnhui, powdered and extracted with95%methanol for three times. Then the resulting mixture,which was obtained via vacuum distillation unit, was diluted with water, and extracted with ethylacetate and dichloromethane. The alkaloid constituents and non-alkaloid constituents wereobtained from Piper nigrum L. From Piper nigrum L.3compounds were isolated. The structuresof these compounds were elucidated by means of spectroscopic methods including1D-NMR(1H-NMR,13C-NMR) and2compounds were identified by spectral analysis:1:methyl piperate2:corydalineAbout1%compands are methyl piperate. This pepper is taken from the fresh unpeeled blackpepper. As usually there are lots of piperine followed by the pepper alkali. Piperine has goodanti-cancer effects. Methyl piperate has a good lipid-lowering effect. The active ingredient in freshunpeeled black pepper was different between sun-baked pepper. When we use the pepper as spiceswe should notice the fresh pepper is different with the sun-backed pepper. Especial low lipidpatients should pay attention to this point.
