Alginate is a polymer which use α-L-guluronate and its C5epimer β-D-mannuronate to form the linear and viscous polysaccharide. The alginate may be thehomopolymeric of guluronate and mannuronate, or polymer of alternately guluronate/mannuronate or random heteropolymeric of guluronate/mannuronate. Due to theunique physical and chemical properties of alginate, alginate has a wide range ofapplications in such aspects as food, chemical industry, medicine, agriculture.Alginate lyase catalyze the degradation of alginate by a β-eliminationmechanism, and form a double bond between the C4and C5of the six-memberedring, finally produce the oligosaccharide mixture which contain uronic acid. The mainsources of alginate lyase were microbes, marine plants and animals, because of thedifferent acting site of the alginate lyase it has different substrate specificity.We isolated a bacterium from the corrupted kelp, strain5-3-2-2-1, andthrough the detection of16S rRNA sequencing and genome to identify the strain. Wedecompose the fermentation broth of the strain5-3-2-2-1to get crude enzyme extract,and determine the determination of alginate lyase activity, the method of drawingstandard curve of protein, finally we choose the appropriate separation methods forseparation and purification of alginate lyase.
