In this study, based on analysis the characteristics and anti-inflammatory mechanisms of several commonly used anti-inflammatory drugs, such as naproxen, ketoprofen and flufenamic acid, we synthesized three novel compounds used these three drugs as the raw materials by esterification reaction with borneol. There were naproxen borneol ester, ketoprofen borneol ester and flufenamic acid borneol ester. These compounds were characterized by MS, NMR and HPLC. In this study, the Cayman COX inhibitor screening assay kit was used to test these three compounds cyclooxygenase inhibition performance in vitro. The results showed that the compounds had the same inhibition performance of cyclooxygenase with their bulk drugs basically. The transdermal experiment of the compounds in vitro was investigated in this study. The results indicated that these compounds had good percutaneous absorption properties. Live mice were used in animal pain test model. Compared with the original medicines, after smeared on the body surface of mouse in a certain time, the pain response in mice was inhibited significantly by the novel compounds. In addition, anti-tumor trial was also investigated using the compounds, the results showed that the proliferation of tumor cells was inhibited significantly.
