Diabetes is one of the chronic diseases which serious threat to human health, by injecting insulin and taking drags to regulate blood sugar are the main treatment method of diabetes. And alpha inhibitor drugs has become one of the mainstream drug for treating diabetes. The alpha glycosidase inhibitor that is from the natural is being a hot research area in recent years. According to the report, Cordyceps militaris has a good hypoglycemic effect, which the main active ingredient is polysaccaride in Cordyceps militaris.This paper researched the inhibition on alpha glycosidase of Cordyceps militaris polysaccharides, then to explore one of function beverage which can regulate blood sugar level.Cordyceps militaris fruiting body as the main raw material, the basic composition and the main functional component were analyzed. Cordyceps polysaccharide content is21.85%and protein content is19.54%and cordycepin content is1.08%.The yield of the polysaccharide that in Cordyceps militaris fruiting body as the review indicators. The water extraction method, ultrasonic assisted extraction and microwave assisted extraction were studied. Investigating the optimal extraction conditions of each methods and the polysaccharide yield of three kinds of methods respectively are7.31%,8.49%and5.77%, through the single factor and orthogonal experiment. By analysising and comparing get that the highest polysaccharides yield of these method is the ultrasonic assisted extraction. Its extraction technology conditions were as follows:extraction temperature60℃, extraction time60min, extraction.power225W, material liquid ratio1:30(g/mL), then to put in the80%ethanol and let it stand overnight, centrifugal and sed-imentation, freeze drying to get the coarse polysaccharide.Through to establish the enzymatic reaction in vitro and acarbose as the positive control, maltose as the reaction substrate. Getting that the polysaccharide inhibition rate gradually increase with the improvement of relative content which different relative sugar content of cordyceps militaris inhibition on a-glycosidase enzyme. The contrast experiment of different purity of Cordyceps militaris polysaccharides at the same concentration (2mg.mL-1) shows that the purity of polysaccharide is more higher that the inhibition rate of cordyceps militaris on a-glycosidase enzyme is more stronger. Cordyceps crude, alcohol after heavy crude polysaccharide, in addition to the polysaccharide and protein purified polysaccharide of Cordyceps militaris for a-glycosidase enzyme inhibition rate is30.79%,33.33%,36.59%and43.67%respectively. With different concentrations of Cordyceps militaris polysaccharide purified on a-glycosidase enzyme inhibition validation experiment of Cordyceps militaris polysaccharide of yeast source a-glycosidase enzyme with certain concentration dependence effect, its value concentration of IC50is4.22mg/mL. Indicated that the Cordyceps militaris polysaccharide after purified polysaccharide has significant inhibitory effect for a-glycosidase enzymes.The best condition to keep the stability of the beverage is to distribute xanthan gum and CMC by3:2ratio, adding amount is0.1%, the stability coefficient is0.981on the basis of stability coefficient, viscosity and the drink storage experiment. According to the single factor primary experiment, orthogonal experiment and combination with fuzzy sensual ev-aluation get that the best formula of the beverage is adding40%of caterpillar fungus ext-ract juice, xylitol9%and citric acid0.1%.The rheological type of Cordyceps militaris beverages is pseudoplastic fluid through the research of rheological properties which can be used for good description by power rate equation. Its viscosity will rise as the rise of the temperature and Arrhenius equation can w-ell describe the influence of the temperature on viscosity of Cordyceps militaris beverages. Taking the sensory evaluation, microorganism and the colour of beverage as the review ind-icators obtain the best sterilization conditions is to sterilizate at90℃for20min.
