In this study,corn powder was served as raw material through the pretreatment,enzymatic hydrolysis,eparation and purification to produce high F value oligopeptide,itsprocess was mainly discussed.The conclusions are as follows:1.The pretreatment of corn powder was studied.According to a research on singlefactor test and orthogonal test,the optimum process conditions of corn powder was asfollows:universal mill,after a100mesh sieve,pasting under90℃for one hour,alphaamylase enzyme solution under adding0.5%,pH value5.0,time1.5h,temperature85℃.2.The processing conditions of enzymolysis were studied and the conclusion ofselecting2709alkaline protease as incision enzyme,flavor protease as excision enzymewas reached.On the basis of single factor,the experimental conditions were optimizedthrough the ternary regression orthogonal test,during which the ratio of2709alkalineprotease and flavor protease,pH value,temperature were choosed as factors and the degreeof hydrolysis as index.It is concluded that the optimal process of double enzymecomposite enzyme solution for:substrate concentration9%,pH value9.6,the ratio of2709alkaline protease and flavor protease13:12(total amount of enzyme was8%),temperature50℃,time4h.3.The separation and purification processing conditions were studied and the bestadsorbent adsorption effect was activated carbon among732cation exchange resin,activated carbon,and HP20macroporous resin.On the basis of single factor,theexperimental conditions were optimized through orthogonal test,during which activatedcarbon dosage,pH value,time were selected as factors,absorbance ratio at220nm and280nm as index.The optimal process for adsorption of aromatic amino acids was asfollows:0.5%dosage of activated carbon,pH value3.0,temperature35℃,adsorption time60min.The best working pressure of removing polypeptide was0.2MPa with membrane efficiency as indicator.The best working pressure of desalination was0.4MPa withdesalination rate as indicator.4.The molecular weight of corn peptide was analyzed using high pressure liquidchromatography,the results show that corn peptide molecular weight under1000Daltonreached98.01%,and the molecular weight of between190and190Dalton reached73.73%,the molecular weight of less than190Dalton reached5.99%,the product obtainedwas oligopeptide,the high value was33.28,and whose protein content was91.4%,moisture content was4.5%,ash content was5.7%,all reached the national standard....
