With the rapid development of the economy and the society, people has increasing demand for food, the pursuit of health and the high quality of life has become a problem.So to strengthen the supervision of the food industry is becoming more and more necessary.At present, a dozen additives in wine standard have be mentioned, but because wine merchants blind pursue aromas, palate,colour and lustre, more illegal additives may be added in wine. According to the results of investigation on wine, it found that in recent years, there is a big problem in the use of additives in wine. Therefore, in order to regulate domestic wine market, it is neccessry to improve detection techniques. The purpose of the study is to establish an effective method for additives measured simultaneously. The method of HPLC had easy operation and high performance liquid chromatography (HPLC)-mass spectrometry (MS) is an analytical chemistry technique that combines the powerful separation capabilities of HPLC with high sensitive detection capabilities of MS. Therefore in recent years, HPLC-MS as a new detection technology is being developed rapidly.As industrial, agricultural and domestic water contained excessive amounts of nitrogen and phosphorus have been discharged into the water, this will lead to accelerated eutrophication process. Algal toxins (especially microcystin) produced by algae has attracted people’s attention, because it has been proven to be a strong carcinogenic agent induced human liver cancer. MC is not only a threat to human health through drinking water, and can be gathered in other aquatic animals and plants, furthermore, can threaten human health through the food chain. In order to prevent and eliminate MCs hazards to humans and animals and plants, and reduce the occurrence of similar incidents of poisoning, the only way is to monitor and control MC limits in various water bodies, marine flora and fauna. For quantitative detection on MC, the high-performance liquid chromatography (HPLC) is considered the most classic of reliable technology. This study aims to establish a convenient and feasible, accurate and reliable, and quickly analyze four kinds of microcystin by HPLC method at the same time. And using the normalization method for microcystin each isomer determines purity. Then a simple and effective method was developed for the separation of four isomers. The work provides an important basis for the subsequent preparation of microcystin toxin standard substance and the detection of microcystin toxin in water quality and organisms.
