In recently years, drug delivery system has been extensively studied and widely used because drug delivery system can increase the utilization of drugs, reduce the dose of drugs and reduce the side effects. As a medical polymer, Eudragit polymers are widely used in the area of pharmaceutical.In this study, colon-targeting polymer Eudragit S100particles are prepared by Gas Antisolvent (GAS) process and Aerosol Solvent Extraction System (ASES) process. The effects of operating conditions such as temperature, pressure, concentration and solvent on the influence of particle morphology and particle size of the polymer were investgated.Base on the study of pure ploymer, prednisolone was used as a model drug, and Eudragit S100was used as a coating material. In the ASES process, prednisolone and Eudragit S100were coprecipitated by using acetone and methanol as the solvent and supercritical CO2as the antisolvent. SEM data showed how the various process variables including pressure, temperature, prednisolone concentration and solution flow rate make effects on morphology and particle size. The efficiency of nanoparticles loading prednisolone was investigated by a UV spectrophotometer. The vitro release profiles exhibited no prednisolone release at gastric (pH=1.2), while continuous release of drug was observed at colonic pH (pH=7.4).This study shows prednisolone loaded Eudragit S100nanoparticles has colon-targeted characteristics.
