Electrospinning technique is an effective method for the preparation of nanofibers, nanofibers membrane exhibit many extraordinary properties including a large surface area, porosity, and easy to separation and recycling rich structure, durable and so on.Laccase is a copper-containing polyphenols oxidase enzyme proteins restored, with a broad substrate specificity and good stability, as a kind of efficient, green catalyst bleached pulp biological, chemical synthesis, food production, environmental pollution, biological and immunological testing and other areas has important application value.Especially in the aspect of environmental protection, laccase is often used to degrade phenolic compounds in water, purification, and soil environmental monitoring and other fields, has been widely studied. Using electrostatic spinning preparation of nanometer fiber membrane as the carrier of the immobilized enzyme, will help to improve the amount of immobilized enzyme, and is conducive to the diffusion of substrates to the enzyme active site and a product outside the reaction zone, the catalytic efficiency of the immobilized enzyme can be significantly improved.In this study of choose poly (methyl methacrylate)(PMMA) and P-cyclodextrin (β-CD) as raw material blend, prepared using the electrostatic spinning PMMA/β-CD nanofibers membrane.By studying the results, under the condition of feed rate was0.2ml/h, the applied voltage was18kV and tip-to-collector distance was set at15cm.that can gained reproducible and bead-free nanofibers. β-CD may be added to adjust the morphology of the nanofibers, β-CD in the amorphous state uniformly distributed in the composite nanofibers increasing amount of β-CD, PMMA/β-CD nanofibers wettability, electrical conductivity also will be increased significantly.Laccase was immobilize on PMMA/β-CD composite nanofibers by physical adsorption. Study of pH, temperature, time et al on the lacquer immobilized enzyme immobilization. Experimental results show that when the fixed condition of laccase at40℃, buffered at pH=5.0was immobilized6h, the maximum specific activity of the immobilized enzyme. Through analysis of the free enzyme and immobilized enzyme catalytic reaction kinetics, it is concluded that immobilized laccase compared with the most suitable for a free laccase enzyme reaction temperature increase, The optimum enzymatic reaction pH also increases, and the change of pH curve widened obviously. Laccase immobilized enzyme kinetic parameters Km rise, Vmax decreased, which decreased enzyme activity is consistent and reflects the good biological affinity between the carrier and the immobilized enzyme.The storage stability was significantly improved and the reusability was very high after laccase were immobilized onto the surface of PMMA/β-CD composite nanofibers membranes.
