Based on the discussion of DHA microcapsule powder, studying the stability of DHA microcapsule preparation technology in infant milk powder, And put forward the new method of LC-MS in the determination of DHA in milk, the main research contents results are as follows.1 The stability of DHA microcapsule powderThe effects of stability of microencapsulated powder DHA was comparative analysised from aerobic and vacuum packaging/nitrogen packaging, temperature and heat/cold/freezing and so on. Research results show that:the light, aerobic conditions, such as high temperature storage conditions promoting DHA microcapsule oxidation. DHA microcapsule powder with foil bag, vacuum packing room place 2 months, POV values change from the initial value 1.7meg/kg to 3.01 meg/kg, DHA reduced to 94.0% retention rate is about the stability of oil powder is good.2 The preparation research of adding DHA Microcapsule into infant formulaThis study was prepared by adding DHA spray drying microencapsulation techniques of infant formula study, The results showed that adding DHA to infant formula microcapsules optimum conditions:homogenization pressure of 40MPa, homogeneous 40-45℃, spray drying, inlet temperature 170℃, the air temperature is 70℃; Under these conditions, the microcapsules prepared by adding milk DHA in the product warranty period of product quality is extremely stable, does not have any adverse reaction, DHA content in the product warranty period and no significant changes.3 The use of LC-MS determination of DHA in infant formulaOn the base ofresearch in the comparative study of different methods (enzymatic walls broken walls,ultrasonic extraction method)and different extraction methods (Acid hydrolysis method, alkali ether, chloroform-methanol method) for infant formula of preprocessing method on the determination of DHA, discussessing the influence by high performance liquid chromatography/mass spectrometry infant formula of new determination method of study. The results show that:broken by enzymatic extraction of oil and the alkaline ether combined pretreatment can extract DHA from milk; high performance liquid chromatography/mass spectrometry in the DHA content of milk powder conditions were:methanol:water 80:20,5min to 23min gradient, milk samples based DHA to achieve the desired separation, the method has the mobile phase is simple, short, simple and easy to grasp, accurate, reproducible. Method has the advantage of established without chromatography mass spectrometry to time constraints and to achieve repeat analysis of samples.
