This paper optimizes the technology of extraction and buccal tablets of polysaccharides which is extracted from Polygonatumodoratum (Mill.) Druc. It provides an experimental and theory foundation for a further application of Polygonatum odoratum (Mill.) Druc. The following researches have been carried out:The polysaccharides of Polygonatum odoratum (Mill.)Druc is determined via the UV-vis spectrophotometry method at 286 nm with fructose as the standard. The structure of polysaccharides of Polygonatum odoratum (Mill.) Druc is identified by IR. The results show that the characteristic absorption peaks of polysaccharide at 3437.91cm-1, 1741.91cm-1,2927.61 cm-1,1248.12cm-1,1025.20cm-1and so on. Through the elements analysis, the comparison of constructure of elements in polysaccharides of Polygonatum odoratum (Mill.) Druc before and after the membrane separation has been made. The comparison results show that each content of element has been decreased in polysaccharides of Polygonatum odoratum (Mill.) Druc. However, the decreasing amplitude of C and H elements is great larger than that of N and S elements. This phenomenon proves that this membrane separation method can remove micromolecule carbohydrate, purify polysaccharides of Polygonatum odoratum (Mill.) Druc, and preserve its structure and biological activity.By RP-HPLC analysis, the results show that the major components of polysaccharide of Polygonatum odoratum (Mill.) Druc are fructose and a few malmose, glucose and xycose, but there is no galaetose. The ability to remove hydroxyl radical and superoxide anion is tested. The results show that Polygonatum odoratum (Mill.) Druc can remove hydroxyl radical and superoxide anion. Compared with VC, it has a weaker affection in removing hydroxyl radical, but has a stronger affection in removing superoxide anion. The ability of removing hydroxyl radical and superoxide anion get stronger when the concentration of Polygonatum odoratum (Mill.) Druc increases. The single factor experiments, center composite design have been used to optimize the processes of extracting polysacchafides of Polygonatum odoratum (Mill.) Druc and manufacturing buccal tablets of Polygonatum odoratum (Mill.) Druc polysaccharides. The optimum water extraction conditions of polysaccharides from Polygonatum odoratum (Mill.) Druc is obtained as following:ratio of solid and liquid is 1:18.5, temperature is 76℃, the duration of time is 111 min with 2 repetitions. The yield of polysaccharides of Polygonatum odoratum (Mill.) Druc is 8.76%. The optimum technology of manufacturing buccal tablets of Polygonatum odoratum (Mill.) Druc polysaccharides is obtained as following:the mass of microcrystalline cellulose is 3.04 g, the mass of malmose is 3.75 g, ratio of aspartame and citric acid is 5:4.4. Theory comprehensive score of buccal tablets of Polygonatum odoratum (Mill.) Druc is the highest, up to 94.56.
