Polygonatum odoratum used both as food and folk medicine,is a naturally abundant plant.Its broad function includes nourishing yin and moisturize the lung,clearing heat and expelling damp,dilation of the coronary arteriae,reducing blood cholesterol,improve myocardial ischemia,relieve atherosclerosis plaque,anti-aging,and anti-cancer effect.Thus investigation on comprehensive utilization of Polygonatum odoratum is of great importance.This report covers the extraction and purification of polysaccharide and total saponins of Polygonatum odoratum,which will provide base for further exploration of Polygonatum odoratum and its related products.Based on the single-factor experiments,response surface methodology and orthogonal test were employed to optimize the extraction and decolourization conditions of polysaccharide from Polygonatum odoratum.The optimum ultrasonic extraction conditions were as follows: ultrasonic power of 359 W,ultrasonic time of 21 min and solid-liquid ratio of 1:20(g/m L).Under the optimized conditions,the extraction rate of polysaccharide reached 5.17%,which was in good agreement with the predicted value.The optimum decolourization conditions were determined as follows: amount of active carbon of 1.5 g,p H of 7,shaking time of 30 min under 70 o C,the decolourization rate was 28.44% and residual rate of polysaccharide was97.21%.Based on the single-factor experiments,taking sarsasapogenin as the standard,response surface methodology was employed to optimize the different parameters,which affected the extraction rate of total saponins,and determine the technology conditions for extraction of total saponins from Polygonatum odoratum.The optimum ultrasonic-assisted extraction conditions were as follows: ultrasonic power of 354 W,ultrasonic time of 31 min and solid-liquid ratio of 1:27(g/m L).Under these optimized conditions,the extraction rate of total saponins reached 4.72%,which was in good agreement with the predicted value.The optimum pressurized-solvent method extraction conditions were as follows: extract pressure at 2.2MPa,material-liquid ratio at 1:22(g/m L)and extracting time at 33 min.under the optimum extracting condition the average content of total saponins in extracts from Polygonatum odoratum was 4.52%.The optimum extraction conditions as follows: microwave power was 388 W,microwave time was 41 min,ethanol concentration was 50%,the extraction rate of total saponins was 4.47% under the optimum extraction conditions.Using the method of UV spectrophotometry,the adsorption rate as main index,the amount of D-101 resin,adsorption temperature,adsorption time,sample concentration to carry out a single factor experimental study.Based on the adsorption experiments,the best conditions for the adsorption were determined: the amount of resin is 3.0 g;the best adsorption time is 4 h;sample concentration is 84.68 ?g/m L.Then in the desorption process,the effect of eluent concentration and amount was investigated using the single factor experiment,the optimum condition: eluant concentration is 85%,the best elution amount is 20 m L.After optimization,the final adsorption rate can reach 42.18%,the desorption rate can reach 91.70%.
