Polyporus umbellatus(Pers.) Fries,as a kind of medicinal fungi, has the immune, nootropic anti-aging, anti-tumor, antioxidant effect.Polysaccharides are natural macromolecular compound, participate in and maintain a variety of physiological functions of life. This paper aims to test the separation and purification of Polyporus polysaccharide, explore their biological activity, provide a basis for the development and utilization of Polyporus polysaccharide.We take the Polyporus umbellatus raw as the experimental materials.Test extraction, purification, antibacterial activity, antioxidant activity of inquiry. The experimental results are as follows:1.The sporophore powder was extracted by ultrasonic assisted, first were extracted with 78% ethanol, ratio of liquid to material 8:1(mL/g), ultrasonic extraction temperature 60 ℃, two hours extraction, of 6 times, extraction after ethanol extraction liquid and recovering solvent to obtain alcohol extract 655 g, the extract rate of 3.6%; and using deionized waterultrasonic extraction, solid-liquid ratio 1:4, extracted 2 hours at temperature 60 ℃, 5 times. Recovery of water after alcohol precipitation were 360.5 g of sugar.sugar yield of 1.98%.2.Polyporus Polysaccharide after removing protein, pigment removal, dialysis, through DEAE-52 cellulose gel chromatography to obtain with the water elution and 0.1 mol/L NaCl elution. And then puting 0.1 mol/L NaCl elution partially purified by gel G-100, were obtained PPS1-1 and PPS1-3-1 two molecular weight uniformity of Polyporusus Bellatus, respectively: the molecular weight of 18268 Da(250.5 mg) and 25203 Da(290 mg). IR shows that PPS1-1 and PPS1-3-1 polysaccharides are characteristic absorption.UV scanning results show that PPS1-1 and PPS1-3-1 are free of nucleic acids and proteins, The analysis shows it be binding protein.Specific rotation PUPS PPS1-1 and PPS1-3-1 were 57.132 ° and 76.388 °; Monosaccharide analysis Results: PPS1-1 mainly composed of xylose and an unknown composition, anotherwise also contains a small amount of rhamnose, fructose, and mannose. PPS1-3-1 mainly consists of fructose, xylose and an unknown sugars, containing a small amount of rhamnose and mannose. Polyporus Polysaccharide PPS1-1 as white powder, PPS1-3-1 as white floc.3.Crude Polyporus Polysaccharide, PPS1-1 and PPS1-3-1 antibacterial activity were screened by filter paper method. The experimental results show that the molecular weight uniform Polyporus Polysaccharide and Polysaccharide PPS1-1 and PPS1-3-1 have no antibacterial activity.4. Crude Polyporus Polysaccharide, PPS1-1 and PPS1-3-1 for DPPH· and hydroxyl radical(?OH) has certain scavenging ability, and DPPH·removal ability is better. with the increasing concentration within a certain range, showing a good amount effect relationship. Polyporus polysaccharide PPS1-1 and PPS1-3-1 on DPPH· and hydroxyl radical(OH) scavenging activity was significantly less than that of crude Polyporus polysaccharide.Overall: Classical water extraction and alcohol precipitation to obtain PUPS, through isolated and purified a different molecular weight and homogeneity PUPS PPS1-1 and PPS1-3-1, for the subsequent structure solution, structure-activity relationship studies provide a certain basis.
