Polyporus umbellatus is prepared from the dried sclerotia of P.umbellatus(Pers.)Fries(Polyporaceae).It is one of the most important traditional medicines and has many kinds of biological activity,such as diuretic,anti-tumor and antioxidant.Polysaccharide,organic acid,steroid and protein are its mainly chemical constituents.So far,the studies of Polyporus umbellatus have been focused maily on polysaccharide and steroid.But the chemical constituents are complex and there are lots of undiscovered small molecules in Polyporus umbellatus,which is a serious restrict on its development.Therefore,studying on the chemical constituents of Polyporus umbellatus is very necessary and significant.The ethyl acetate part of ethyl alcohol extract of P.umbellatus has anti-cancer and antibacterial activities.The experiment is to research on the ethyl acetate part to supply some theoretical basis for the development and utilization of the chemical constituents of P.umbellatus.In this experiment,the ethyl acetate part of the 78% ethyl alcohol extract of P.umbellatus were extracted,isolated and tested of bioactivity.The results are as follows:1.The ethyl acetate extract(2 mg/m L)could inhibit the proliferation of HeLa cells in some degree.Besides,we found that it also had a good inhibition on Alternaria solani.When the extract concentration was 50?g/m L,the inhibition rate was up to 51%,which was stronger than that of the positive control of carbendazim(the inhibition rate is 6%).The experiment also evaluated the in vitro antifungal properties of the ethyl acetate extract against two fungi(Staphylococcus aureus and Escherichia coli).It showed inhibition activity,but the effect was less than that of the positive control of fosfomycin sodium(the drug loading is 10?g).2.There were 18 compounds separated from the ethyl acetate fraction of the 78%ethyl alcohol extract of P.umbellatus sclerotia through solid-liquid extraction,column a-bsorption chromatography unde rnormal pressure,recrystallization et al.11 compounds a-mong them were characterized by ways such as NMR spectra,HRMS.The ethyl acetate extract could inhibit the HeLa cells,Alternaria solani,Staphyloc occus aureus and Escherichia coli.But it had no activity on Fusarium graminearum,and Fusarium solani.We used compounds that weights were greater than 10 mg testing the antibacterial activity.These tested compounds had no antibacterial activity on th ose five bacteria.Of the 11 compounds,3(ZY1,ZY2,ZY15)of them were steroidal compounds;6 fatty acids(ZY3,ZY4,ZY10,ZY14,ZY16,ZY17)were isolated from Polyporus umbellatus(Pers.)for the first time.1(ZY11)was containingnitrogen;1(Z Y13)was polyphenolic compound.
