Study Of The Extraction Of Northeast Hericium Polysaccharide And Its Function In Relieving Physical Fatigue

Northeast Hericium is growing at a low temperature, and growing in a long time, whose fruiting body is more closely. Northeast Hericium is rich in nutrition and tastes more plump and delicious. The polysaccharide as the active ingredient in Northeast Hericium can anti ulcer and tumor, but it has been reported that rarely the polysaccharide in Northeast Hericium can relieve physical fatigue has been reported rarely. Because of busy life, "over the model worker" cannot fully escape from the pressure. Therefore, relieving physical fatigue and fighting fatigue become a hot topic in sports medicine and health medicine. This paper studied the influence of the Northeast Hericium polysaccharide in relieving physical fatigue effect in mice, which provided a reference for activity studing of reliveing physical fatigue and health food development.(1) The active ingredients in the fruiting bodies of Northeast Hericium were analysed qualitatively:The active ingredients in the fruiting bodies of Northeast Hericium were analysed qualitatively by physical and chemical methods. It contained flavonoids, organic acids, alkaloids, phenolic substances, fat, amino acids, polysaccharides, saponins, terpenes and other active substances.(2) The research of the extraction and purification of Northeast Hericium polysaccharide:The Northeast Hericium polysaccharide were extracted by water extraction and alcohol precipitation, on the basis of single factor experiments, and made the response surface optimization tests, then analyzed the optimum extraction parameters of Northeast Hericium polysaccharide as follows:extraction temperature was 100℃, extraction time was 5h, liquid ratio was 25:1 (mL/g), and the extraction rate was 4.53%. Compared with TCA method, using Sevage method to remove protein from Northeast Hericium polysaccharide was better. Protein removal rate was 99.1%. Northeast Hericium polysaccharide was obtained with different volume fraction of ethanol. According to HPLC chromatograms and total antioxidant capacity of the experimental results, the volume fraction of 80% ethanol was identified as the best volume fraction of ethanol.(3) The study of classification of Northeast Hericium polysaccharide:The crude polysaccharide was separated by ion-exchange chromatography into neutral and acidic polysaccharide. Then different concentration of NaCl water solution was used to further classifiy and six fractions was obtained. A single symmetrical peak was obtained by gel column chromatography. The uniform six fractions, in which NHPA-4 and NHPA-5 displayed more than one map dispersed in Sepharose CL-6B, and no clear single symmetrical peak; NHPA-1-1, NHPA-1-2, NHPA- 2-1, NHPA-2-2, NHPA-2-3 single symmetrical peak had a broad distribution of total sugars and uronic acid assay, which showed less acidity polysaccharide content; NHPA-3-1’s peak was a single symmetrical, and concentrated acidic polysaccharide. Therefore, NHPA-3-1 was obtained by combining DEAE- cellulose and Sepharose CL-6B, and a yield was 3%. Full wavelength UV spectrum scan results showed the higher levels of purity points. NHPA-3-1 monosaccharide composition of mannose:glucuronide:glucose:galactose=2.2:21.82: 68.37:7.97.(4) The study of Northeast Hericium polysaccharide in relieving physical fatigue functions:Northeast Hericium polysaccharide in relieving physical fatigue function was evaluated by using animal loading swimming experiments. The inbred mice were randomly divided into three groups with normal saline negative dose group.The mice were fed NHPA-3-1 (gavage volume of 4mL/20g body weight of mice) in low-dose group and high dose groups, with respectively,3mg/20g body weight and 9mg/20g body in 30 days The animals weight increased, and had no deaths; high dose group compared to the negative control group, the mice loading swimming time was significantly longer,and vivo mouse liver glycogen reserves were more original amount, and mouse blood lactate and serum urea nitrogen were less amount, which indicated the Northeast Hericium polysaccharide with a certain relieving physical effects of fatigue.
