Tiller onion belongs to the Liliaceous Allium plant.Tiller onion has been experts found that at least six kinds of anticancer anti-ageing ingredients lipid-lowering effect,and it has a good therapeutic effect on the breast,prostate,brain tumor and other diseases.In this study,Tiller onion was used to research enzymatic extraction of optimal parameters of crude polysaccharides and through the optimization technology of enzymatic assisted ultrasonic extraction of Tiller onion water-soluble polysaccharides by response surface method.Then the water-soluble polysaccharides were separated and purified.Finally,the mice model was used to detect the effect of polysaccharides on reducing blood lipid.The main contents in this paper were concluded as follow:i.The method of water bath extraction was used to extract Tiller onion water-soluble polysaccharides and single factor test on enzyme dose,extraction time,extraction temperature and ratio of material to liquid was used to research on the extraction effect of Tiller onion polysaccharide.And then through the orthogonal test to determine the optimum parameters of water extraction of Tiller onion polysaccharide:the enzyme dose is 2%,the material is at 1:20,the temperature is 55?,and the time is 2 hours.Under the best process,the extraction yield can reach 8.30%.ii.The ultrasonic method was used to examine response surface p H value,ultrasonic waves,ultrasonic power effects on extraction yield of polysaccharide from Allium.Obtained in a dry extract of optimal conditions for: p H values of 5,ultrasonic power 200 W,ultrasonic time was 40 min,Tiller onion in the yield of Water-soluble polysaccharide was 15.2% ? Compared with ultrasonic and microwave assistant methods,the yield of Tiller Onion's Water-soluble polysaccharide increased by 3.58%.iii.The resin was used to purify the polysaccharides from onion.With the decolorization rate and the retention rate of polysaccharide as the test index,Single factor experiments on static and dynamic adsorption were carried out respectively in order to preliminarily optimize the Tiller Onion water-soluble polysaccharide purification process.The results show that the selection of AB-8 resin is more suitable.Static adsorption: the appropriate adsorption time is about 2h,and the suitable amount of resin is 1.5g.Dynamic adsorption: the concentration of the upper sample is 2000mg/L,p H is 5,and the temperature of the upper sample is 40?,and under this condition,the decolorization rate was 84.6% and the retention rate of polysaccharide was 81.2%.iv.The hyperlipidemia model mice were intragastric administration of Tiller Onion polysaccharide to identify its hypolipidemic effect.The Tiller Onion polysaccharide dose groups compared with model group,the organ coefficients were decreased which show that Tiller Onion polysaccharides have different effects on organs.The mice which were continuously gavaged to a certain dose of polysaccharide can inhibit the content of TC,TG,LDL-C in serum of mice,that shows Tiller Onion polysaccharide can effectively prevent hyperlipidemia rats serum TC and TG content,namely in reducing serum TC,TG,LDL-C content significantly.And the Tiller Onion polysaccharide can effectively maintain the HDL-C content in serum of hyperlipidemia mice,that is,it plays an important role in improving the content of serum HDL-C.Tiller Onion polysaccharide can effectively prevent arteriosclerosis,which has obvious effect on reducing the arteriosclerosis index(AI).
